S‐Endo 1, a pan‐endothelial monoclonal antibody recognizing a novel human endothelial antigen

Abstract: A murine monoclonal antibody (mAb) S-Endo 1 has been produced to detect circulating endothelial cells detached from blood vessels in pathological conditions. We have demonstrated that the associated-antigen (S-Endo 1 Ag) was highly expressed on human vascular structure irrespective of tissue origin or vessel caliber. Its expression was not restricted to endothelium, since it was also detected at low level on smooth muscle cells, stroma cells and follicular dendritic cells. But its absence on hematopoietic cell… Show more

“…CD146 is ubiquitously expressed on endothelial cells [28] and melanoma cells [29]. A correlation with poor prognosis has recently been found in breast cancer; of 635 primary breast tumors, 7% were CD146-positive, and CD146 expression was associated with poor prognosis [12].…”

“…Sickle cell disease is a vascular disease that causes tissue ischemia and exaggerated endothelial turnover, both of which are characterized by evident endothelial injury and regeneration [34]. In different patient groups, the number of ECs/mL of peripheral blood may vary between zero and 1,500; most studies found 1 to 100 circulating ECs/mL in healthy people [34][35][36][37][38][39][40]. We found much higher numbers of circulating ECs in both sickle cell patients and controls, probably due to methodological differences in sample preparation and cell quantification.…”

“…Typically, 100 μl of bead suspension is then non-covalently coated with 10 μg/ml CD-146 antibody (Novus Biologicals, Littleton, CO, USA). CD-146 is expressed almost exclusively in mature endothelial cells with the exception of some tumour cell lines [23,24].…”
