Rules of engagement for base excision repair in chromatin

Abstract: Most of the DNA in eukaryotes is packaged in tandemly arrayed nucleosomes that, together with numerous DNA- and nucleosome-associated enzymes and regulatory factors, make up chromatin. Chromatin modifying and remodeling agents help regulate access to selected DNA segments in chromatin, thereby facilitating transcription and DNA replication and repair. Studies of nucleotide excision repair (NER), single strand break repair (SSBR), and the homology-directed (HDR) and non-homologous end-joining (NHEJ) double stra… Show more

“…Several factors, such as the cyclin/cyclin-dependent kinase (cyclin/CDK) complex, closely regulate all the processes of each phase (15). If cell proliferation is disturbed by infection with the influenza A virus, the host cells are arrested in the G0̸G1 phase (16) and if the DNA damage is not repaired, the cells enter the apoptotic process (17).…”

“…Further, therapeutic approaches are considered to target epigenetic machinery components [57,58]. By dissecting out BER response and epigenetic landscape cross talk, there are appreciable suggestions that BER pathway influenced by chromatin disruption and remodeling [59]. The DNA methylation is known as a stable epigenetic modification.…”

“…Little is known about how DNA glycosylases can detect base lesions with high accuracy; this task is extraordinarily challenging because the differences between modified and natural bases are very subtle and the glycosylases are often present at low levels. A threedimensional search termed 'facilitated diffusion' is associated with DNA glycosylase recognition (Zharkov and Grollman, 2005), and chromatin based-mechanisms are also believed to impact this step (Odell et al, 2013;Rodriguez et al, 2015). For more sensitive detection of irregularities, selected bases are flipped out of the helix into a selective pocket for substrate interrogation in the subsequent step.…”
