RT-QuIC Assays in Humans … and Animals.

Abstract: Prion diseases are neurodegenerative diseases affecting both humans and animal species. The phenotypic spectrum is broad and includes Creutzfeldt-Jakob disease (CJD) and its variant zoonotic form (vCJD) in humans, while in animals, scrapie of sheep and goats, bovine spongiform encephalopathy and chronic wasting disease of deer, elk and moose are naturally occurring forms. Transmission and pathogenesis appear causally linked to the misfolding of the normal form of the prion protein (PrP C ) into disease associa… Show more

“…Thioflavin T, a benzothiazole dye added to the reaction mixture, exhibits enhanced fluorescence and a characteristic red shift of its emission spectrum after binding to the amplified αSyn rec aggregates. The resulting fluorescence signals indicate the occurrence and progress of aggregate amplification, i.e., the effect of αSyn seeding activity present in the test sample, and can be recorded in real time [ 9 , 11 , 26 ].…”

“…Most animal studies using asyn SAA methods are confined to prion disease research (reviewed by Collins and Sarros, 2016 and Atarashi, 2022 ). Recent successful use of animal tissue for the conversion of human asyn substrate also strengthens the position of the asyn SAA as a robust research tool.…”
