RSUME, a Small RWD-Containing Protein, Enhances SUMO Conjugation and Stabilizes HIF-1α during Hypoxia

Abstract: SUMO conjugation to proteins is involved in the regulation of diverse cellular functions. We have identified a protein, RWD-containing sumoylation enhancer (RSUME), that enhances overall SUMO-1, -2, and -3 conjugation by interacting with the SUMO conjugase Ubc9. RSUME increases noncovalent binding of SUMO-1 to Ubc9 and enhances Ubc9 thioester formation and SUMO polymerization. RSUME enhances the sumoylation of IkB in vitro and in cultured cells, leading to an inhibition of NF-kB transcriptional activity. RSUME… Show more

“…41 SUMOylation of HIF-1a has also been described by several groups, however the reported outcomes of this modification are varied. For example, in a number of studies, SUMOylation of HIF-1a has been suggested to both increase HIF-1a stability and transcriptional activity, 57,58 but in others, it has been suggested to decrease activity and enhance VHLmediated ubiquitination 59,60 Transcriptional Cofactors…”

“…In addition, one of the important functions of NF-κB in tumour cells or cells targeted by carcinogenic agents is promoting cell survival, by inducing the expression of anti-apoptotic genes (such as BCL2). There is also accumulating evidence of interconnections and compensatory pathways between the NF-κB and HIF1α systems [28][29][30] , linking innate immunity to the response to hypoxia.…”

“…However, defining the relationship between NF-kB and HIF-1a has proven elusive. Using in vitro systems, it was reported that HIF-1a activates NF-kB 9 , that NF-kB controls HIF-1a transcription 10 and that HIF-1a activation may be concurrent with inhibition of NF-kB 11 . Here we show, with the use of mice lacking IKK-b in different cell types, that NF-kB is a critical transcriptional activator of HIF-1a and that basal NF-kB activity is required for HIF-1a protein accumulation under hypoxia in cultured cells and in the liver and brain of hypoxic animals.…”
