Routes to Bioactive Hydrophilic Polymers

Abstract: ABSTRACT:The reactions employed for coupling anti-platelet and other drugs to hydroxylic polymers are described. The drugs include the prostaglandin analogue 5-(6-carboxyhexyl)-1-(3-cyclohexyl-3-hydroxypropyl) hydantoin (BW 245C) and the phosphodiesterase inhibitors dipyridamole and theophylline. Polymers studied were poly(vinyl alcohol), dextran, and poly(ethylene glycol), and coupling was effected either with the aid of halo-isocyanates or by a two-stage process using carbonyl diimidazole. These novel reacti… Show more

“…These side-chains behave as spacers separating the bioactive units from the polymer backbonesthe spacer length is often an important feature influencing bioactivity in such systems. The spacer may be hydrophilic (CH 2CH 2 0 )n or hydrophobic (CH 2CH 2 )m' We have found significant effects on the molar activity of coupled BW 245C produced by variations in spacer length and hydrophilicity in polymers based on N-vinyl pyrrolidone (R = ot:J ) studied in aqueous solution (11,14).…”

“…A relevant example is the vasodilator dipyridamole (Persantin, Boehringer, Mannheim, Germany) [10) which may be esterified with methacrylic acid to yield the monomer [11) (DIMA). This ester provides a useful method for coupling dipyridamole to polymers.…”
