Endoplasmic reticulum (ER) stress plays a pivotal role in the pathogenesis of asthma. The present study aimed to investigate the reducing or suppressing effects of crocin in ovalbumin (OVA)-sensitized mice on ER stress markers.Mice were divided into six groups (n = 5 per group) including control, OVAsensitized (OVA), OVA-treated crocin (OVA-Cr25, OVA-Cr50, and OVA-Cr100 mg/kg), and OVA-treated dexamethasone (1 mg/kg), (OVA-Dexa) groups. Animals 5 later groups were sensitized to OVA and the treatment groups received intraperitoneally crocin/dexamethasone in the last 5 days of the model. At the end of the study, lung tissue was evaluated for airway inflammation, caspase 12 and CHOP protein levels, and expression of ER stress markers using realtime-PCR. Sensitization with OVA significantly caused airway inflammation and induction of ER stress in mice compared to the control group based on the elevated inflammatory cells and ER stress markers in the lung tissue. Treatment with crocin and dexamethasone reduced airway inflammation and suppressed ER stress markers. Interestingly, in the OVA-Cr100 group, the suppressive effects on ER stress apoptotic markers were comparable to the OVA-Dexa group. The results suggest that crocin mediates maladaptive ER stress conditions possibly by creating adaptive ER stress status and driving protein folding correctly.

The consumption of nuts and extra-virgin olive oil has been associated with suppression of inflammatory pathways that contribute to atherosclerosis, but its role on the modulation of the inflammatory profile in patients with established coronary artery disease (CAD) is unclear. The aim of this study was to evaluate the effects of adding pecan nuts or extra-virgin olive oil to a healthy diet on inflammatory markers in patients with stable CAD. In this randomized clinical trial, 204 patients were enrolled to three study groups: 67 to control group (CG: healthy diet), 68 to pecan nuts group (PNG: 30g/day of pecans + healthy diet) and 69 to extra-virgin olive oil group (OOG: 30ml/day of extra-virgin olive oil + healthy diet). High-sensitivity C-reactive protein (hs-CRP, in mg/L), fibrinogen (mg/dL), interleukins (IL) 2, 4, 6, 10 (pg/mL) and interferon-γ (IFN-γ, in pg/mL), IL-6/IL-10, IL-2/IL-4 and INF-γ/IL-4 ratios were evaluated at baseline and after the follow-up (12 weeks). As main results, after adjustment for sex, statin used and relative body weight variation, there were no differences between groups regarding inflammatory markers at the end of the study. IL-6 levels (primary outcome) were reduced in 12 weeks when compared to baseline in all study groups [CG: difference: −0.593 (SE = 0.159) pg/dL; PNG: difference: −0.335 (SE = 0.143) pg/dL; OOG: IL-6 difference: −0.325 (SE = 0.143) pg/dL]. In conclusion, there was no significant effect of including pecan nuts or extra virgin olive oil to a healthy diet on inflammatory markers in individuals with CAD.

Objectives Acute respiratory distress syndrome (ARDS) is occurred by disruption of alveolar-capillary membrane and associated with an inflammation process. In the present study, the effect of arnafant surfactant on RDS in rats was compared to curosurf. Methods RDS was induced in male Wister rats by surfactant washout using 10 times lung lavage with 3 ml saline at 37oC through an intra-tracheal cannula after anesthetized with intrapritonal administration of 60 mg thiopental Na while animals were ventilated. Then chest was opened and stepwise increasing volume of air was administered and withdrawer from the lung while lung pressure was monitored and lung compliance was determined using lung volume-pressure curve. Total and differential white blood cells (WBC) as well as oxidative stress markers in the blood and bronco-alveolar lavage (BALF) were also examined. The study was performed in 4 groups (n=10 in each group) of rats including control and RDS, both traded with saline, RDS treated with arnafant and RDS treated with curosurf (2.5 ml contained 6.25 mg surfactant for both drugs).Results The value of lung compliance in RDS group was significantly reduced compared to control group (P<0.001). However, in RDS group treated with arnafant, lung compliance was significantly increased compared to both untreated and to curosurf treated RDS groups (P<0.001 for both cases). Total and differential WBC counts in the blood of RDS group were significantly increased compared to the control group but in RDS group treated with arnafant, total WBC, and lymphocyte counts were significanty decreased compared to untreated RDS group (P<0.01-P<0.001). In the BALF of RDS group treated with curosurf, total and differential WBC were significantly increased but in RDS group treated with arnafant, only total WBC, eosinophil and lymphocyte counts were significanty increased compared to the control group (P<0.01-P<0.001). In the BALF, lung tissue and serum of untreated RDS group, all oxidative stress markers were significantly changed compared to the control group but in arnafant treated RDS group all markers and in curosurf treated RDS group, only thiol level and CAT activity in the BALF as well as MDA and thiol levels both in the lung tissue and serum were significanty improved compared to untreated RDS group (P<0.05-P<0.001).Conclusion Treatment of RDS rats with arnafant significantly improved lung compliance as well as lung and systemic inflammation and oxidative stress and the effect of this surfactant drug was significantly higher than curosurf.
