Comparative Quantitative Studies on the Microvasculature of the Heart of a Highly Selected Meat-Type and a Wild-Type Turkey Line

In this study the macroscopic and microscopic structure of the heart of a fast growing, meat-type turkey line (British United turkeys BUT Big 6) and a wild-type turkey line (Canadian Wild turkey) were compared. At 8 and 16 weeks of age, 10 birds of each genotype and sex were sampled. The body mass and heart mass of the meat-type turkey both increased at a faster rate than those of the wild-type turkey. However in both turkey lines, the relative heart mass decreased slightly with age, the decrease was statistically significant only in the male turkeys. Furthermore meat-type turkeys had a significantly (p < 0.01) lower relative heart mass and relative thickness of the left ventricle compared to the wild-type turkeys of the same age. The wild-type turkeys showed no significant change in the size of cardiomyocytes (cross sectional area and diameter) from 8 weeks to 16 weeks. In contrast, the size of cardiomyocytes increased significantly (p < 0.001) with age in the meat-type turkeys. The number of capillaries in the left ventricular wall increased significantly (p < 0.001) in wild-type turkeys from 2351 per mm2 at the age of 8 weeks to 2843 per mm2 at 16 weeks. However, in the meat-type turkeys there were no significant changes, capillary numbers being 2989 per mm2 at age 8 weeks and 2915 per mm2 at age 16 weeks. Correspondingly the area occupied by capillaries in the myocardium increased in wild-type turkeys from 8.59% at the age of 8 weeks to 9.15% at 16 weeks, whereas in meat-type turkeys this area decreased from 10.4% at 8 weeks to 9.95% at 16 weeks. Our results indicate a mismatch in development between body mass and heart mass and a compromised cardiac capillary density and architecture in the meat-type turkeys in comparison to the wild-type turkeys.

Preservation of original tissue dimensions is an essential prerequisite for morphometric studies. Shrinkage occurring during tissue processing for histology may severely influence the appearance of structures seen under the microscope and stereological calculations. Therefore, shrinkage has to be avoided so that estimates obtained by application of unbiased stereology are indeed unbiased. The present study investigates the alterations of tissue dimensions of mouse lung samples during processing for histology. Different fixatives as well as embedding protocols are considered. Mouse lungs were fixed by instillation of either 4% formalin or a mixture of 1.5% glutaraldehyde/1.5% formaldehyde. Tissue blocks were sampled according to principles of stereology for embedding in paraffin, glycol methacrylate without treatment with osmium tetroxide and uranyl acetate, and glycol methacrylate including treatment with osmium tetroxide and uranyl acetate before dehydration. Shrinkage was investigated by stereological measurements of dimensional changes of tissue cut faces. Results show a shrinkage of the cut face areas of roughly 40% per lung during paraffin embedding, 30% during "simple" glycol methacrylate embedding, and <3% during osmium tetroxide/uranyl acetate/glycol methacrylate embedding. Furthermore, the superiority of the glutaraldehyde-containing fixative regarding shrinkage is demonstrated. In conclusion, the use of a glutaraldehyde-containing fixative and embedding in glycol methacrylate with previous treatment of the samples with osmium tetroxide and uranyl acetate before dehydration is recommended for stereological studies of the mouse lung.

Impact of Autonomic Tonus Typological Features on the Duodenum Structure of Chickens

The goal of this research is to show the relationship between the duodenum morphology of laying hens (Gallus domesticus) of ISA Brown cross and typological peculiarities of autonomic tonus in the poultry organism. To achieve the goal a group of adult chickens (aged one year) was investigated to determine an aggregate tonus of their autonomic centers based on the study of heart rate variability (variation pulsometry). According to the results all the poultry was divided into two groups: sympatheticotonic chickens (hereinafter referred to as ST chickens) and sympathetico-normotonic chickens (hereinafter referred to as ST-NT chickens) The first group of poultry is characterized by a clear predominance of sympathetic centers tonus while within the second group there is a slightly increased tonus of parasympathetic centers. Using a set of various morphological methods a comparative analysis of the duodenum structure in these groups was made. The analysis revealed that ST chickens unlike ST-NT chickens are characterized by a higher value of the duodenum absolute length (by 1.6 cm) and its relative length (by 1.4 %) as well as lower values of its internal perimeter (by 0, 25 cm). Apart from this, an increase of absorption surface of the duodenum mucous membrane is observed in ST-NT chickens. Increase in villus height (by 143.7 micrometers) and their quantity (by 61.5) per 1sm 2 contribute greatly to it. In the duodenum muscle membrane the thickness of both layers has larger values under a distinct sympathetic tonus in ST-NT chickens. In a circular layer difference between poultry groups reaches 21 micrometers while in a longitudinal it approaches 4.9 micrometers. Investigating a connective tissue it was found that the relative area of its fibers near the mucosal crypts of ST chickens is larger by 0.21 % than that of the ST-NT chicken. In the muscle layer area a superiority of ST chickens according to this indicator reduces to 0.15%.
