Role of vimentin in modulating immune cell apoptosis and inflammatory responses in sepsis

Abstract: New diagnostic biomarkers or therapeutic targets for sepsis have substantial significance for critical care medicine. In this study, 192 differentially expressed proteins were selected through iTRAQ. Based on cluster analysis of protein expression dynamics and protein-protein interactions, hemopexin, vimentin, and heat shock protein 90 were selected for further investigation. It was demonstrated that serum vimentin (VIM) levels were significantly increased in patients with sepsis and septic shock compared to c… Show more

“…HIF1α participates in the regulation of a plethora of cellular events such as metabolism of ROS trough the regulation of SOD2 39 , the regulation of cytoskeleton trough VIM type III lament, which also participates in in ammation 40 , and PLAUR which activates plasminogen and activates a cascade of extracellular proteases 41 . Interestingly, the expression of this gene could be used as a predictor of severe respiratory failure 42 which is consistent with our results.…”

“…The effects of quercetin, at a safe working concentration, on the cell were identified by fluorescence observation (Ndozangue-Touriguine et al, 2008;Su et al, 2019). GS cells (1 × 10 4 ) were cultured in 35-mm glass-bottom dishes (Cellvis, catalog number D35-14-1-N) at 28°C for 24 h and then incubated with quercetin (50 μg/ml) for 24 h at 28°C.…”

“…Vimentin may prevent lymphocyte apoptosis and be anti-inflammatory and, thus, may be a useful new target in sepsis [82]. Vimentin is one of the main constituents of the intermediate filament proteins that maintain cell shape, in part, by stabilizing the interactions of the cytoskeleton and perhaps more importantly by helping cells resist damage including apoptosis.…”

“…Hemopexin was one of 56 proteins identified in plasma, finally ranking fourth in a protein network of 12 based upon interactions with eight other proteins in the network (defined as "betweenness centrality"). Thus, HPX has been linked in a co-expression protein network with vimentin in patients diagnosed with sepsis and septic shock [82].…”
