Role of Versican and ADAMTS-1 in Polycystic Ovary Syndrome

Abstract: Objective:ADAMTS-1 is a matrix metalloproteinase which cleaves versican in the cumulus oocyte complex under the effect of luteinizing hormone surge in the periovulatory period. Altered levels may have a role in the pathogenesis of polycystic ovary syndrome (PCOS). We aimed to determine the serum versican and ADAMTS-1 (a disintegrin and metalloproteinase with thrombospondin motif-1) levels in PCOS patients and compare the results with healthy controls.Methods:Thirty-eight patients with PCOS and forty healthy co… Show more

“…Since we could not identify from the literature any versican concentration typical for trauma patients, we assumed (analogous to syndecan-1) a 10-fold increase of its plasma concentration. Since plasma versican concentrations in healthy subjects are 95 ng/ml on average, versican (product code 230–00833, RayBiotech, Norcross, USA) was added to achieve a final concentration of 950 ng/ml [ 16 ]. An additional sample was prepared using the concomitant application of all substances in their respective final concentrations outlined above.…”

“…These differences may be due to the sample sizes and experimental conditions. Nonetheless, there are studies did not show any significant difference in the expression of ADAMTS1 between the two types of patients (Karakose et al, 2016;Özler et al, 2017), which might be due to the detection methods and ethnicity of the patients.…”

“…Serum levels of lnc‐ADAMTS1–1:1, lnc‐SAMD14–5:3, lnc‐COL1A1–5:1, and lnc‐GULP1–2:1 may also serve as potential markers of ovarian follicle reserve or POI. In addition, the expression of lnc‐ADAMTS1–1:1 was correlated with its predicted target gene ADAMTS1, which is important in normal follicular development (Ozler et al, ; Shozu et al, ); and this correlation was specific to ovarian cortical tissues, but not to other human tissues. Therefore, we speculate that lnc‐ADAMTS1–1:1 may also be a valuable POI serum marker.…”
