Role of the endothelium and nitric oxide synthases in modulating superoxide formation induced by endotoxin and cytokines in porcine pulmonary arteries

Abstract: Background: The interactive roles of cytokines, endotoxins, superoxide (O 2 v -) and nitric oxide (NO) in the pathogenesis of adult respiratory distress syndrome (ARDS) have not been fully elucidated. The effects of tumour necrosis factor-α (TNF-α), interleukin 1α (IL-1α), and lipopolysaccharide (LPS) and the role of NO and the endothelium in mediating O 2 v -formation were therefore investigated in intact porcine pulmonary arteries in vitro. Methods: Intrapulmonary artery (PA) segments were obtained from Whit… Show more

“…It is possible that NO or NOx are protective during ALI. NO scavenges oxygen free radicals that are generated during oxidative stress (4,33). NO vasodilates the microcirculation, which allows for increased perfusion of tissue beds (33).…”

“…The measurement of O 2 ·Ϫ released by cells was performed by detection of ferricytochrome c reduction. 7 Thus, after incubation for 16 hours, pulmonary arterial cells were rinsed 3 times in PBS to remove drugs and stimulators and preequilibrated in DMEM without phenol red for 10 minutes at 37°C in a 95% air-5% CO 2 incubator. Then 20 mol/L horseradish cytochrome c (Sigma Chemical Co) with or without 500 U/mL copper-zinc superoxide dismutase (SOD; Sigma Chemical Co) was added to the segments or cells and incubated at 37°C in a 95% air-5% CO 2 incubator for 1 hour.…”

“…5 In turn, TNF-␣ and ILs and lipopolysaccharide (LPS) upregulate enzymes that generate O 2 ·Ϫ in cultured vascular tissues, in particular, NADPH oxidase. 6,7 A selfperpetuating inflammatory cascade ensues.…”

“…Nicotinamide adenine dinucleotide phosphate oxidase is upregulated by many platelets and leukocytes factors such as entothelin-1, leukotrienes and serotonin. 69 O 2 À promotes VSMC proliferation and migration and upregulates MMPs (for a review, see Jeremy et al 68 ). O 2 À also reacts with NO to produce reactive nitrogen species reducing NO bioavailability, which is itself associated with vein graft disease as mentioned above.…”
