Role of Prostaglandin E2 and Leukotriene B4 in Skin Reaction Induced by Transdermal Application of Propranolol.

Abstract: Dermal application of propranolol (PRL) induced formation of erythema and edema, and pseudoeosinophil infiltration in epidermis and dermis at the application site in guinea pigs. We investigated the production of prostaglandin E2 (PGE2) and leukotriene B4 (LTB4) at the application site of PRL and the role of these inflammatory chemical mediators in the occurrence of the skin reactions. PGE2 was found to be produced at the application site slightly after the accumulation of PRL released from the adhesive bandag… Show more

“…But many enhancers cause skin irritation and PHCl has an irritating effect on skin itself. It was reported (Kobayashi et al, 2000) that skin irritation with propranolol correlated to the amount that penetrated through the skin, and the same phenomena was observed by us with increased PHCl transdermal flux.…”

“…Amongst the other antihypertensives, propranolol, a drug with desirable hydrophilic-lipophilic balance, has created high expectations. Enormous data has been generated for this drug as it is also used as a model molecule for mechanistic studies and system development [48][49][50][51][52] . However, initial studies carried out to evaluate www.ijpsonline.com permeability showed that the drug induces inflammatory reactions 53 .…”

“…Dexamethasone is a synthetic glucocorticoid [14] widely used in inflammatory condition [15] and to enhance the normal immune response [16]. It is used as a therapeutic agent in alcohol withdrawal syndrome [17], cerebral edema [18], congenital adrenal hyperplasia [14], nausea, and vomiting, especially associated with a high dose of anticancer agents [19], high altitude disorder [18], cerebral malaria, opportunistic mycobacterial infections, respiratory disorders, skin disorders [14], and rheumatism [20].…”
