Role of parathymic lymph nodes in metastatic tumor development

Abstract: Parathymic lymph nodes as potential sites of tumor progression have been neglected in humans. We have established a rat renal capsule-parathymic lymph node model to study in vivo metastasis. Epithelial liver carcinoma (HeDe) and mesenchymal mesoblastic nephroma (NeDe) cell lines have been established after inducing chemical carcinogenesis in newborn Fisher 344 inbred rats by N-nitrosodimethylamine. Implanting the exact number of tumor cells (HeDe, NeDe) under the renal capsule allowed the standardization and t… Show more

“…Using this approach, we could locate a distant parathymic lymph node metastasis after intraperitoneal lymphoma injection with intravenous Ga-67 citrate administration followed by SPECT/MRI and ex vivo CLI. This metastatic location is in agreement with earlier demonstration of DLBCL lymphoma spreading following intraperitoneal injection [44]. However, further optimization strategies are needed to explore the full potential of this novel approach.…”

“…An impairment of its function causes lymphedema and ascites [57]. Another important fact is that liver is one of the most frequent sites of tumor formation [64]. Tumor staging prognosis and therapeutic approaches for cancer are most often based on the extent of involvement of regional lymph nodes [59].…”

“…After adaptation to the microenvironment cancer cells start to divide 44. Recently the lymphoid-hematogen aspect of metastasis formation prevails, but the spread of broken away tumor cells from the peritoneal cavity to the thoracic lymph nodes is more in the favor of a lymphatic drainage 45, 46.…”

“…Our metastatic rat tumor model clarified that due to delayed angiogenesis, the inner part of primary tumor necrotized. The arterial blood pressure and the lack of venous backflow channeled blood and tumor cells to the interstitial fluid 39, 45, 46.…”
