We aimed to study lymphoma diagnostics by Cerenkov luminescence imaging (CLI). We monitored the dissemination of a spontaneous high-grade mouse lymphoma (Bc.DLFL1) in early stage; advanced stage; and terminal stage with in vivo 2-deoxy-2-[18F]fluoro-D-glucose ([18F]FDG) positron emission tomography (PET) / magnetic resonance imaging (MRI) and 67Ga-citrate single photon emission computed tomography (SPECT) / MRI. In vivo imaging was combined with ex vivo high resolution CLI. The use of CLI with Fluorine-18 and Gallium-67 to select infiltrated lymph nodes for tumor staging pathology was thus tested. At advanced stage, [18F]FDG PET/MRI plus ex vivo CLI allowed accurate detection of [18F]FDG accumulation in lymphoma-infiltrated tissues. At terminal stage we detected tumorous lymph nodes with SPECT/MRI and we could report the Cerenkov light emission of 67Ga. CLI with 67Ga-citrate revealed lymphoma accumulation in distant lymph node locations, unnoticeable with only MRI. Flow cytometry and immunohistochemistry confirmed these imaging results. Our study promotes the combined use of PET and CLI in preclinical studies and clinical practice. Heterogeneous [18F]FDG distribution in lymph nodes detected at sampling surgery has implications for tissue pathology processing and could direct therapy. The results with 67Ga also point to the opportunities to further apply suitable SPECT radiopharmaceuticals for CLI.

Importance Rat Liver Morphology and Vasculature in Surgical Research

BackgroundThe laboratory rat is one of the most popular experimental models for the experimental surgery of the liver. The objective of this study was to investigate the morphometric parameters, physiological data, differences in configuration of liver lobes, biliary system, and vasculature (arteries, veins, and lymphatic vessels) of the liver in laboratory rats. In addition, this study supports the anatomic literature and identified similarities and differences with human and other mammals.Material/MethodsForty laboratory rats were dissected to prepare corrosion casts of vascular system specimens (n=20), determine the lymph vessels and lymph nodes (n=10), and for macroscopic anatomical dissection (n=10) of the rat liver. The results are listed in percentages. The anatomical nomenclature of the liver morphology, its arteries, veins, lymph nodes, and lymphatic vessels are in accordance with Nomina Anatomica Veterinaria.ResultsWe found many variations in origin, direction, and division of the arterial, venous, and lymphatic systems in rat livers, and found differences in morphometric parameters compared to results reported by other authors. The portal vein was formed by 4 tributaries in 23%, by 3 branches in 64%, and by 2 tributaries in 13%. The liver lymph was drained to the 2 different lymph nodes. The nomenclature and morphological characteristics of the rat liver vary among authors.ConclusionsOur results may be useful for the planing of experimental surgery and for cooperation with other investigation methods to help fight liver diseases in human populations.

The aim of this study is to select among potential tumor models that could be suitable to follow the metastatic spead of tumor cells. 18FDG-PET tumor diagnostic test has been adapted to investigate tumor growth in vivo in local and metastatic rat models. Materials and Methods. The expression of glucose transporters was traced by immunohistological analysis, followed by the uptake of 18FDG and visualized by MiniPET scanner. After s.c. administration of hepatocarcinoma (He/De) cells intensive local tumor growth and 18FDG uptake were measured. Results: Whole body 18FDG-PET imaging supported by histological analysis have shown that subcutaneously growing tumors did not project metastases to other sites from the injected area. To avoid local tumor formation i.v. injection was chosen, but did not improve the safety of tumor cell administration. Tumor formation after i.v. injection took a longer time than after s.c. administration. Tumors upon i.v. generation were smaller and detectable in liver and lung, but not in other organs or tissues. iii) Subrenally implanted He/De cells spread from the retroperitoneal primary tumor of the kidney to thoracal paratymic lymph nodes (PTNs). The spread from primary site to metastatic tumors in PTNs was confirmed by post mortem surgery and histological examinations. Conclusion: Among the three methods applied: a) Local s.c. administration of tumor cells generated local tumors unsuitable to study metastasis. b) Intravenous administration causing unpredicatable location of tumor formation is not regarded a reliable metastatic tumor model. c) Subreanal implantation model proved to be a suitable model to follow the metastatic process in rats.
