Nitric oxide (NO) has been recently reported to play an important role in the rewarding effects of addictive drugs. The regional NO signaling in the brain, however, is not completely clear. Here, we studied the effects of insular NO signaling on the expression of morphine-induced conditioned place preference (CPP). Insular microinjection of the NO inhibitors N-nitro L-arginine methyl ester (L-NAME) and 7-nitroindazole reduced the expression of morphine-induced CPP. The NO donor molsidomine, in contrast, reversed L-NAME-induced reduction of CPP expression. These results suggest that insular NO signaling is involved in the expression of morphine-CPP.

N-methyl-D-aspartate receptor 2B subunit (NR2B) has an important role in the development of conditioned place preference (CPP) and psychostimulant abuse. Rhynchophylline is presently used to treat central nervous systems diseases and has a non-competitive antagonistic effect on NMDA receptors. In this study, amphetamine was administered in rats (2 mg/kg, s.c., once each day for 4 consecutive days), during which they were treated with rhynchophylline (60 mg/kg, i.p., once each day for the next 3 days). NR2B mRNA and protein expression were examined by in situ hybridization and immunohistochemistry. CPP was induced by amphetamine (2 mg/kg, s.c.) by 4th day in rats. Rhynchophylline effectively reversed the expression of amphetamine-induced CPP and itself did not produce a CPP. Amphetamine-CPP rats showed a significantly increased NR2B mRNA and protein expression in medial prefrontal cortex and hippocampal CA1 areas as compared to the control group. Rhynchophylline reversed NR2B mRNA and protein levels induced by amphetamine but rhynchophylline by itself had no effect on NR2B expression in control rats. These results indicate that rhynchophylline inhibits the expression of amphetamine-induced rewarding effect, and this action might be related to down-regulation of NR2B expression in medial prefrontal cortex and hippocampal CA1 area.

Nitric oxide (NO) plays obligatory roles as an important intercellular messenger in the control of physiological functions and it also participates in pathophysiological interventions. This labile, gaseous molecule is also involved in mechanisms underlying the beneficial and untoward actions of therapeutic agents. Endogenous NO is formed by endothelial and neurogenic NO synthases that are constitutively present mainly in the endothelium and nervous system, respectively, and is induced by lipopolysaccharides or cytokines mainly in mitochondria, glial cells, and vascular smooth muscle cells. NO modulates the effects of morphine on processes involving the central nervous system, such as learning, memory, convulsion, thermoregulation, and penile erection. This molecule is also involved in the modification of morphine actions on the cardiovascular, digestive, and respiratory systems. Morphine regulates NO bioavailability in various organs. NO formed by inducible NO synthase participates in some morphine actions in the immune system. Information concerning interactions between NO and morphine and other opioids in a variety of organs and tissues is quite useful in establishing new strategies for minimizing the noxious and unintended reactions that are frequently encountered during analgesic therapy.
