Role of Nicotine Dependence in the Association between the Dopamine Receptor Gene DRD3 and Major Depressive Disorder

Abstract: BackgroundThe aims of this study were to analyze associations of dopamine receptor genes (DRD1-5) with Major Depressive Disorder (MDD) and nicotine dependence (ND), and to investigate whether ND moderates genetic influences on MDD.MethodsThe sample was ascertained from the Finnish Twin Cohort. Twin pairs concordant for smoking history were recruited along with their family members, as part of the multisite Nicotine Addiction Genetics consortium. Genetic association analyses were based on 1428 adults. Total of … Show more

“…In psychiatric samples, smokers have greater impulsivity, aggression, suicidal ideation and suicide attempts and, among those with depression, lower indices of serotonin function (Malone et al 2003 ). Nicotine exposure moderates the effect of dopamine receptor gene polymorphisms on major depressive disorder incidence (Korhonen et al 2014 ). Additionally, tobacco smoke contains multiple other compounds which may contribute, whose effect on central nervous system function in humans is poorly known (US Department of Health and Human Services, 2014 ).…”

“…Other symptoms of MDD include poor concentration and memory, as well as thoughts of death or suicide [7]. MDD is also associated with an increased risk of cardiovascular disease, osteoporosis and diabetes [8,9], and is linked to risk factors such as smoking and alcohol dependence [10,11].…”
