Role of NF-κB in the Antiproliferative Effect of Endothelin-1 and Tumor Necrosis Factor-α in Human Hepatic Stellate Cells

Abstract: During chronic liver diseases, hepatic stellate cells (HSC) acquire an activated myofibroblast-like phenotype and proliferate and synthesize fibrosis components. Endothelin-1 (ET-1), which inhibited the growth of human myofibroblastic HSC, increased the formation of two NF-B DNA binding complexes; this effect was also observed with tumor necrosis factor-␣ (TNF-␣). The complexes were identified as the p50/p50 and p50/p65 NF-B dimers. Activation of NF-B was associated with the degradation of the inhibitory prote… Show more

“…It should be noted that NS-398 decreased basal PGE 2 production, in agreement with our previous data showing that COX-2 is constitutively expressed in hMF (see Fig. 5A) and accounts for basal PGE 2 production in these cells (6). Therefore, these results demonstrate that S1P stimulates an early COX-2-dependent production of PGE 2 , leading to cAMP synthesis.…”

“…Most studies of liver fibrosis reported until now have considered that activated HSC was the principal fibrogenic cell type contributing to ECM accumulation. 58 However, other cell types such as portal fibroblasts may, after activation, acquire fibrogenic potential. 59,60 In our study, a heterogeneous population of hIHFs was isolated from human livers using collagenase perfusion, because most (but not all) of these cells store vitamin A in the same way as stellate cells, when quiescent.…”

“…Glucocorticoids are one class of agents that sometimes, but not always, inhibits myofibroblast formation and proliferation (32,33), perhaps by blockade of TGF-␤ production (34). Other therapies to blunt fibrosis are under study, but none has been shown to be efficacious and nontoxic (35).…”
