Tachykinin NK3 receptor (NK3R) is a g-protein coupled receptor that is heavily expressed by magnocellular neurons of the paraventricular nucleus of the hypothalamus (PVN). Osmotic challenge is reported to activate NK3R expressed by magnocellular neurons and cause the NK3R to be internalized to the cytoplasm and perhaps the cell nucleus. In this study we show using immunoelectron microscopy that isolated nuclei from neurons in the PVN of osmotic challenged animals show a robust labeling for the NK3R. NK3R immunoreactivity was detected by Western blot in isolated nuclei of PVN neurons following the 2 M NaCl injection. No nuclear NK3R immunoreactivity was detected in control animals. NK3R antibody specificity was confirmed by small interfering (SI) RNA technology. This study establishes that the NK3R is trafficked to the nucleus of PVN neurons following a peripheral osmotic challenge. The release of these hormones is influenced by a number of transmitters and magnocellular neurons express a number of receptors. Noteworthy is the tachykinin, NK3 receptor (NK3R) which is heavily expressed by magnocellular neurons of the supraoptic (SON) and paraventricular nucleus (PVN) of the hypothalamus (Ding et al., 1999). NK3R expressed by magnocellular neurons appears to have a functional role in the release of vasopressin and oxytocin. Stimulation of NK3R results in an increase in Ca ++ conductance (Buell et al., 1992), induction of c-Fos expression in magnocellular neurons, and causes the systemic release of both hormones (Ding et al., 1999;Smith and Flynn, 2000;Bealer and Flynn, 2003). Furthermore, the release of the endogenous ligand, presumably neurokinin B (NKB), and binding to the NK3R results in the internalization of the peptide-receptor complex (Chawala et al., 1997;Colin et al., 2002;Lessard et al., 2004;Johnson et al., 2004) that can be tracked immunohistochemically. The expression patterns of Tac2 and NK3 mRNA and of pep2, the neurokinin B (NKB) precursor protein, were compared in rats and mice. Pep2 immunoreactivity was observed in fibers, terminals, and perikarya in the brains of both species, but the number of NKB-immunoreactive cells was generally smaller in mice than in the corresponding nuclei in rats. Congruent distribution patterns of Tac2 mRNA and NKB were found in many nuclei of the thalamus and hypothalamus (habenula, anterodorsal nucleus, preoptic area, arcuate nucleus, paraventricular nucleus). However, mice expressed Tac2 mRNA neither in the hippocampus nor in the nucleus of the lateral olfactory tract, in contrast to rats. Accordingly, mice showed no NKB in the projection areas of these nuclei, such as the olfactory tubercle, whereas a clear NKB signal was present in rat tissues. Surprisingly, we found nearly identical NK3 mRNA expression patterns in both species, despite the species differences in NKB expression. Thus, although the expression patterns of Tac2 and NKB are similar in rats and mice, noteworthy differences exist. Our results have important implications for the interpretation of behavioral results concerning the NKB/NK3 system in these species.

Background The neuropeptide oxytocin (OT) is crucial in several conditions, such as lactation, parturition, mother-infant interaction, and psychosocial function. Moreover, OT may be involved in the regulation of eating behaviors. Methods This review briefly summarizes data concerning the role of OT in eating behaviors. Appropriate keywords and medical subject headings were identified and searched for in PubMed/MEDLINE. References of original articles and reviews were screened, examined, and selected. Results Hypothalamic OT-secreting neurons project to different cerebral areas controlling eating behaviors, such as the amygdala, area postrema, nucleus of the solitary tract, and dorsal motor nucleus of the vagus nerve. Intracerebral/ventricular OT administration decreases food intake and body weight in wild and genetically obese rats. OT may alter food intake and the quality of meals, especially carbohydrates and sweets, in humans. Discussion OT may play a role in the pathophysiology of eating disorders with potential therapeutic perspectives. In obese patients and those with certain eating disorders, such as bulimia nervosa or binge/compulsive eating, OT may reduce appetite and caloric consumption. Conversely, OT administered to patients with anorexia nervosa may paradoxically stimulate appetite, possibly by lowering anxiety which usually complicates the management of these patients. Nevertheless, OT administration (e.g., intranasal route) is not always associated with clinical benefit, probably because intranasally administered OT fails to achieve therapeutic intracerebral levels of the hormone. Conclusion OT administration could play a therapeutic role in managing eating disorders and disordered eating. However, specific studies are needed to clarify this issue with regard to dose-finding and route and administration time.
