“…AChE is a mediator of cholinergic signaling that has also been associated with RAS signal transduction. It may be a downstream target of RAS signaling in PC12 cells [40,41]; on the other hand AChE may induce RAS activity in smooth muscle cells [42]. It is currently not known whether similar connections exist in colon epithelial cells.…”

“…Resting tension was readjusted, if necessary, before commencing the experiments. Relaxation in response to relaxant was expressed as % of phenylephrine (1 AM)-induced tone (0.32 T 0.05 g) (a sustained arterial contraction which is 90% of maximum contraction produced by 3 AM phenylephrine) (Choy et al, 2002), and 100% relaxation was considered when active tone returned to baseline level. Manoeuvres such as administration of N G -nitro-l-arginine methyl-ester (l-NAME), iberiotoxin, 4-aminopyridine and endothelium denudation resulted in an enhanced phenylephrine (1 AM)-induced tone (33.6 T 4.9% increase), concentration of phenylephrine used was therefore adjusted accordingly (0.4 -0.7 AM) in order to have a similar magnitude of active tone developed.…”

“…Only one concentration of individual drug was tested in each cell. Where stated, concentration of inhibitors employed in this study was the reported effective concentration of individual agent, based on our previous studies (Kwan et al, 1999;Choy et al, 2002;Au et al, 2003) and other group (Zhao et al, 2001), tested on pulmonary and other vascular smooth muscle preparations.…”
