Role of microRNA 146a on the healing of cornea alkali burn treated with mesenchymal stem cells

Abstract: The aim of the present study was to investigate the effect of microRNA 146a (miR146a) on promoting the repair of corneal alkali burn with bone marrow mesenchymal stem cells (MSCs). A total of 24 Sprague-Dawley female rats were divided into a normal group (Control), a normal MSC treatment group (Normal MSCs), an miR146a knockout MSC treatment group (miR146a-low MSCs) and an miR146a high-expression MSC treatment group (miR146a-high MSCs) according to the random number table. Quantitative polymerase chain reactio… Show more

“…MiRNAs, a subclass of 18–24 bp long non-coding single-stranded RNA, are an important regulatory RNA component of EVs. MiRNAs regulate stability as well as post-transcriptional regulation of gene expression by binding to target messenger RNAs (mRNAs) [ 54 ]. These biomolecules participate in various diseases including chronic inflammatory diseases, cancer, heart diseases, and ophthalmic diseases.…”

“…In the case of corneal injury, the role of miRNAs on phenomena such as wound repair, inflammation, maintenance of the corneal/ limbal stem cell pool, lymphangiogenesis, and corneal neovascularization has been studied in the pre-clinical setting. For instance, in a rat model of alkali-induced burn injury, the authors evaluated the therapeutic effect of bone marrow-derived MSCs engineered to over-express miR146a on corneal repair [ 54 ]. A reduction in corneal opacification was observed in vivo with improvement in angiogenesis through the inhibition of VEGF, CD45 and Interferon-γ [ 54 ].…”

“…From this, we can infer that these cells have potential to control the microenvironment during local inflammation, and are candidates for allogeneic cell-based therapies. There have been several studies investigating the use of MSCs from other tissue (bone marrow or adipose tissue) in treating corneal disease to differing success[16-19]. The use of MSCs from tissues other than the cornea has shown limitations for corneal disease models.…”
