Role of Matrix Metalloproteinase-7 in Colorectal Adenomas

Abstract: Matrix metalloproteinases (MMPs) degregade and remodel the extracellular matrix. They are known to be overexpressed as normal mucosa progresses to adenomas and carcinomas. In our prospective study we measured the overexpression of MMP-7 immunohistochemically in various types of colonic adenomas. Although MMP-7 has already been shown to be overexpressed in various types of colonic adenomas, tubular versus villous adenomas had not been further seperated to date. Seventy-six patients had either normal mucosa (n=1… Show more

“…MMP-7 is involved in the progression of human rectal cancer and is a key player in invasion and metastasis, and thus, its assessment may become useful for clinical diagnosis and predicting the prognosis of rectal cancer. 20,21 The correlation of MMP-7 expression with liver metastasis has been also reported. 22 It is synergistically upregulated by the oncogenic transcription factors activator protein (AP)-1 and b-catenin.…”

“…3 Previous study suggests that MMP-7 proteins are mainly expressed in colon cancer cells and strongly associated with tumor progression. 5,6 A high plasma level of leptin was correlated with tumor progression in colon cancer patients. 7À9 Studies suggest that leptin could play crucial roles in different clinical stages of CRC.…”

“…A similar trend was observed for TNF· expression with the highest expression (+80-fold) in the largest most advanced tumors (225-470 mm 3 ). In humans, an increased expression of matrix metalloproteinases (MMP) genes has been associated with early stages of tumor development (15). Here, we observe that MMP-7 gene expression was already enhanced in small polyps (+210-fold) compared to the level found in the mucosal samples.…”
