Role of Lymphocytic Choriomeningitis Virus (LCMV) in Understanding Viral Immunology: Past, Present and Future

Abstract: Lymphocytic choriomeningitis virus (LCMV) is a common infection of rodents first identified over eighty years ago in St. Louis, MO, U.S.A. It is best known for its application in immunological studies. The history of LCMV closely correlates with the development of modern immunology. With the use of LCMV as a model pathogen several key concepts have emerged: Major Histocompatibility Complex (MHC) restriction, T cell memory, persistent infections, T cell exhaustion and the key role of immune pathology in disease… Show more

“…To determine how the abundance of specific Tregs relates to the effectiveness of pathogen clearance, we used the mouse model of LCMV to track Treg response following infection (28). This model was selected because acute or chronic infection can be induced by viral strains that differ only by 2 amino acids that do not affect the relevant T-cell epitopes (29). Thus, the same specific T cells can be examined in distinct infectious contexts and at different time points to provide information on Tregs dynamics following acute or chronic viral exposure.…”

“…Lymphocytic choriomeningitis virus (LCMV) is a widely used disease model for studies of the immune response to viral infection (6). LCMV-Armstrong (Arm), first described in the 1930s after an encephalitis epidemic in St. Louis, Missouri (7), results in an acute viral infection, the rapid resolution of which depends largely on an efficient, virus-specific CD8 + cytotoxic T-cell response.…”

“…There are at least 27 members that are geographically, serologically, and phylogenetically divided into Old World and New World arenaviruses (1). The prototypic lymphocytic choriomeningitis virus (LCMV) infection of mice has long been used as a valuable model with which to study viral persistence and virus-induced immunity and immunopathology (2,3). The arenavirus is composed of a total of four genes on two genomic RNA segments in opposite orientations (1).…”
