Role of Long Non-Coding RNAs in Reprogramming to Induced Pluripotency

Abstract: The generation of induced pluripotent stem cells through somatic cell reprogramming requires a global reorganization of cellular functions. This reorganization occurs in a multi-phased manner and involves a gradual revision of both the epigenome and transcriptome. Recent studies have shown that the large-scale transcriptional changes observed during reprogramming also apply to long non-coding RNAs (lncRNAs), a type of traditionally neglected RNA species that are increasingly viewed as critical regulators of ce… Show more

“…With the development of next-generation sequencing technology, lncRNA has become a hot spot in the field of non-coding RNA research in recent years. LncRNA can regulate gene expression in various ways and is widely involved in the up-regulation or down-regulation of oncogenes or tumor suppressor genes and plays a very important regulatory function in various physiological and pathological activities such as cell differentiation, proliferation, metabolism, apoptosis, and senescence (Ji et al, 2020;Kanwal et al, 2020). The differential expression of lncRNA in malignant tumors and the molecular mechanism of action have been gradually investigated, and more and more lncRNAs have been found to be closely related to the occurrence and development of tumors.…”

“…lincRNA-1526 and lincRNA-1463 are associated with inverse regulation of the activation of the pluripotency genes in reprogramming, while lincRNA-1307 and ladr49/83 can positively regulate the pluripotency genes. LNCPRESS1, Snhg14, Gas5 and Peblr20 are positive regulators of the pluripotency genes in reprogramming and/or embryonic stem cells(ESCs) ( 182 ). Moreover, there are novel approaches to comprehend regulation of the lncRNAs.…”
