Role of LFA-1 and ICAM-1 in Cancer

Abstract: The lymphocyte function-associated antigen-1 (LFA-1) (also CD11a/CD18 and αLβ2), is just one of many integrins in the human body, but its significance derives from its exclusive presence in leukocytes. In this review, we summarize the studies relating LFA-1 and its major ligand ICAM-1 (CD54) with cancer, through the function of lymphocytes and myeloid cells on tumor cells. We consider how LFA-1 mediates the interaction of leukocytes with tumors and the role of ICAM-1 in tumor dynamics, which can be independent… Show more

“…123,124 Expression of ICAM-1 has been documented in most, if not all, cell types in the TME. 125,126 As such, in lung adenocarcinoma, ICAM-1 was shown to be induced in transformed alveolar epithelial cells, ECs, pulmonary lymphocytes, and fibroblast. [127][128][129] In melanoma, thin pre-cancerous lesions expressed a negligent amount of ICAM-1; however, analyses of multilayered melanoma lesions revealed heightened ICAM-1 expression at the basal layer.…”

“…Additionally, we validated the functional clusters for patients #07 and #08 in four independent data sets (Additional file 1: Figure S3). Adhesion molecules, such as integrins, have been implicated in cancer progression [23]. It was reported that recruitment of neutrophils (a granulocytic subset that display similar phenotypic/functional characteristics as PMN-MDSCs) to tumor sites is associated with increased expression of CD11a integrin on neutrophils (also known as LFA-1) [24].…”
