Role of ISG15 protease UBP43 (USP18) in innate immunity to viral infection

Abstract: Innate immune responses provide the host with an early protection barrier against infectious agents, including viruses, and help shape the nature and quality of the subsequent adaptive immune responses of the host. Expression of ISG15 (UCRP), a ubiquitin-like protein, and protein ISGylation are highly increased upon viral infection. We have identified UBP43 (USP18) as an ISG15 deconjugating protease. Protein ISGylation is enhanced in cells deficient in UBP43 (ref. 6). Here we have examined the role of UBP43, e… Show more

“…The importance of USP18 in innate immunity to viral infection was demonstrated using USP18-deficient mice, which were resistant to lymphocytic choriomeningitis virus or vesicular stomatitis virus . 62 Although the exact antiviral activity of the ISG15 pathway remains to be determined, its induction by IKK⑀ may play an important role in the antiviral action of IKK⑀ against HCV.…”

“…To date, only five cellular targets for protein ISGylation have been reported, including Serpin 2a, Jak1, Stat1, phospholipase C␥1, and Erk1/2 (28,44), among possibly hundreds of ISGylated proteins. We recently report that UBP43-deficient mice are resistant to certain virus infections (45). Currently, it is not clear how protein ISGylation contributes to innate immunity.…”

“…Both are important for the control of several viral infections and are crucial mediators of IFNmediated immune responses. 23 GCH1 is the first and rate-limiting enzyme in tetrahydrobiopterin (BH4) biosynthesis. 24 Induction of GCH1 and nitric oxide synthase (NOS) has been suggested to represent two arms of a common pathway required for immunostimulantevoked NO synthesis, and the major function of BH4 may be to support NOS.…”
