“…The intracellular localization of each kinase is associated to the different pathways and stress sensors that culminate in the final event of eIF2 phosphorylation and translation inhibition. The HRI kinase (eIF2K1) is activated in heme deprivation and oxidative stress with ROS release; PKR is the cytoplasmic kinase (eIF2K2) activated by viral infection and foreign dsRNA; PERK/PEK (eIF2K3) is associated to ER and related to UPR pathway, as described before; and GCN2 (eIF2K4) is activated by amino acid starvation and UV irradiation (Bertolotti et al, 2000; Daito et al, 2014; Jain et al, 2014). Those kinases cause the phosphorylation of the eIF2-α at Ser51, which impairs its binding to eIF2B, inhibiting GDP-GTP exchange, which is necessary for Met tRNA position on the 43S ribosomal subunit and initiation of translation (Kedersha et al, 2002).…”

“…Transmitted to humans via mosquitos, it can cause severe and widespread disease, including hemorrhagic fever [139]. The capsid of dengue virus is initially synthesized on the ER and then transiently accumulates on LDs [140, 141].…”

“…Dengue is a common hemorrhagic viral infectious disease (Jain, Chaturvedi, & Jain, 2014). An obvious evidence indicated that inflammatory responses are activated by NLRP3 inflammasomes during dengue virus infection.…”
