Role of interleukin‐7 in bone and T‐cell homeostasis

Abstract: Initially defined as a B-cell growth factor, the pleiotropic nature of interleukin-7 (IL-7) has increasingly become appreciated. Besides its well-known roles in B- and T-cell lymphopoiesis, IL-7 is now known to regulate the homeostasis of both mature T cells and bone cells. In bone, the precise nature of how IL-7 affects osteoclasts and osteoblasts is controversial, since it has a variety of actions in different target cells. These activities are gender-specific and are dependent on whether IL-7 is delivered s… Show more

“…IL-7 is produced by non-hematopoietic stromal and epithelial cells in the thymus, bone marrow, intestine, and lymphoid tissue, 43 but also by dendritic cells 44,45 and B cells. 46 Others have found an inverse correlation between IL-7 and lymphocyte numbers post-HSCT.…”

“…BM contains T cell survival factors, such as IL-7 and IL-15, which are recognized to induce Ag-independent proliferation of memory T cells (25,42,43). Interestingly, we did not find differences in IL-7R␣ nor IL-15R␣ expression in BM vs PB T EM .…”

“…IL-7 was first identified as a pre-B-cell growth factor and plays a role in survival and proliferation of precursor B cells [2]. T-cell homeostasis is significantly affected by IL-7 [1], which also influences overall T-cell differentiation and the conversion of effector T cells into memory T cells by facilitating T-cell survival [3,4].…”
