Role of integrins and their ligands in osteoarthritic cartilage

Abstract: Osteoarthritis (OA) is a degenerative disease, which is characterized by articular cartilage destruction, and mainly affects the older people. The extracellular matrix (ECM) provides a vital cellular environment, and interactions between the cell and ECM are important in regulating many biological processes, including cell growth, differentiation, and survival. However, the pathogenesis of this disease is not fully elucidated, and it cannot be cured totally. Integrins are one of the major receptors in chondroc… Show more

“…This suggests that more integrins ( 18 F-FPRGD 2 uptake) are expressed by structures that show evidence of inflammation (FDG uptake). These observations in vivo are in agreement with our knowledge of the cross talk between integrins and cytokines/inflammatory mediators in vitro in chondrocytes or fibroblast-like synoviocytes [13,26,27,42]. A publication also showed that decrease of synovial inflammation using therapeutic intervention as assessed by a decrease in a composite clinical disease activity index significantly correlated to the decrease observed in 68 Ga-PRGD 2 SUV max [30].…”

“…Integrin a v b 5 is as relevant as integrin a v b 3 in OA pathophysiology [14,16,17,[25][26][27]. One of their ligands is vitronectin and it is noteworthy that its concentration in serum is increased with the Kellgren and Lawrence score [6,26,27].…”

“…Generally, cells attached to FBS protein-coated substrates via integrin α5β1-FN and integrin αvβ3-vitronectin interactions [ 11 , 17 , 18 ]. The integrin expression levels were measured with semi-quantitative RT-PCR to gain understanding of the reason for the differences in the attachment among HT-1080, MDA-MB-231, and HepG2 cells ( Fig 5C ).…”

“…FN is involved in important process such as wound healing, cell adhesion and spreading, growth, migration, and differentiation. 12 The purpose of this study was to analyse the expression patterns of HtrA1 in relationship to FN and to the key growth zones of placenta during the first trimester of gestation.…”
