Albumin (Alb) is the most abundant plasma protein with multiple biological functions, including antioxidative property through its thiol activity. Given that inflammatory bowel disease is associated with a decreased level of Alb and an increased level of Alb oxidation, we asked whether Alb could have a therapeutic effect on colitis. Here we tested this possibility. Bovine serum albumin (BSA) was reductively modified with dithiothreitol (DTT) and administrated via gavage or intraperitoneal injection. Dextran sulfate sodium (DSS)-induced mice colitis was associated with massive oxidative stress, as indicated by the elevated sulfenic acid formation in blood, colon tissues, and feces. Treatment of mice with the reductively modified albumin (r-Alb) attenuated the oxidative stress and reduced local inflammation and tissue injury. These effects of r-Alb were only partially achieved by unmodified Alb and wholly lost after blocking the –SH groups with maleimide. In cultured colon epithelial cells, r-Alb prevented DSS- and H 2 O 2 -induced ROS elevation and barrier dysfunction, preceded by inhibition of sulfenic acid formation and P38 activation. Further analysis revealed that Alb was susceptible to H 2 O 2 -induced oxidation, and it detoxified H 2 O 2 in a –SH group-dependent way. Moreover, Alb reacted with GSH/GSSG via thiol-disulfide exchange and reciprocally regulated the availability of –SH groups. Collectively, our study shows that r-Alb effectively attenuates DSS colitis via –SH group-mediated antioxidative action. Given that the oxidative stress underlies many life-threatening diseases, r-Alb, functioning as a potent antioxidant, could have a wide range of applications.

Wooden or woody breast ( WB ) is a myopathy of the pectoralis major in fast-growing broilers that influences the quality of breast meat and causes an economic loss in the poultry industry. The objective of this study was to evaluate growth and proteome differences between 5 genetic strains of broilers that yield WB and normal breast ( NB ) meat. Eight-week-old broilers were evaluated for the WB myopathy and divided into NB and WB groups. Differential expression of proteins was analyzed using 2-dimensional gel electrophoresis and LC-MS/MS to elucidate the mechanism behind the breast myopathy because of the genetic backgrounds of the birds. The percentages of birds with WB were 61.3, 68.8, 46.9, 45.2, and 87.5% for strains 1-5, respectively, indicating variability in WB myopathy among broiler strains. Birds from strains 1, 3, and 5 in the WB group were heavier than those in the NB group ( P < 0.05). Woody breast meat from all strains were heavier than NB meat ( P < 0.05). Within WB, strain 5 had a greater breast yield than strains 1, 3, and 4 ( P < 0.0001). Woody breast from strains 2, 3, 4, and 5 had a greater breast yield than NB ( P < 0.05). Six proteins were more abundant in NB of strain 5 than those of strains 2, 3, and 4, and these proteins were related to muscle growth, regeneration, contraction, apoptosis, and oxidative stress. Within WB, 14 proteins were differentially expressed between strain 5 and other strains, suggesting high protein synthesis, weak structural integrity, intense contraction, and oxidative stress in strain 5 birds. The differences between WB from strain 3 and strains 1, 2, and 4 were mainly glycolytic. In conclusion, protein profiles of broiler breast differed because of both broiler genetics and the presence of WB myopathy.

Proteins play a central role in cell structure and function. The present study aimed to measure the changes in total protein [TP], albumin [A], globulin [G], and [A]/ [G] ratio in sera and saliva samples of type 2 diabetic patients with and without proliferative diabetic retinopathy (PDR). Meanwhile agarose electrophoresis was used to confirm such changes. Also it aimed to look for the possibility of using saliva as an alternative sample to serum as a diagnostic fluid. The results indicated that a significant increase was observed in serum [G], salivary [TP], [A], and [A]/ [G] ratio, p =0.000, p=0.041, p=0.04, p=0.001 respectively, while there was a significant decrease in serum [A]/ [G] ratio p=0.000 in patients without PDR compared to healthy control. Also, the results showed that there was a significant increase in serum [G], salivary [TP], [A], [G] and [A]/[G] ratio, p =0.000, p=0.000, p=0.000, p=0.000, p=0.037 respectively, while a significant decrease in serum [A]/[G] ratio p=0.000 was observed in patients with PDR compared to patients without PDR. Sera and salivary agarose gel electrophoresis fractionation indicated an increase in [TP] and [G], but the changes were more obvious in the levels of gamma globulin fractions where the higher values were noticed in the patients group with PDR. Sera [A] were slightly decreased while salivary [A] was slightly increased. The most obvious finding was the existence of a positive correlation between the changes in [TP], [A] and [G] of serum and saliva in all studied groups.
