Role of HERG1 potassium channel in both malignant transformation and disease progression in head and neck carcinomas

Abstract: Evidence indicates that human ether à -go-go-related gene 1 (HERG1) voltage-gated potassium channels could represent new valuable membrane therapeutic targets and diagnostic/prognostic biomarkers in various cancers. This study is the first to investigate the expression pattern of HERG1 potassium channel subunit in both primary tumors and precancerous lesions to establish its clinical and biological role during the development and progression of head and neck squamous cell carcinomas. HERG1 protein expression w… Show more

“…Morphologically representative areas were selected from each individual formalin‐fixed paraffin‐embedded tumor block for the construction of a tissue microarray (TMA). Three 1‐mm cylinders were taken to construct TMA blocks, as described previously . A total of 10 TMAs were created, containing 3 tissue cores of each of the 241 oropharyngeal SCCs.…”

“…To avoid cross-contamination, two punches of 2 mm diameter were taken first, using a new, sterile punch (Kai Europe GmbH, Solingen, Germany) for every tissue block, and stored in Eppendorf tubes at room temperature prior to DNA extraction. Subsequently, three 1 mm cylinders were taken to construct TMA blocks, as described previously [10]. A total of 5 TMAs were created, containing three tissue cores of each of the 124 laryngeal and hypopharyngeal carcinomas.…”

“…Inhibition of certain voltage gated Na+ channels reduces the metastatic ability of prostate cancer cells in a rat model by abrogating their motility [96, 98]. Numerous studies suggest that the pharmacological or genetic inhibition of K+ channels can reduce the proliferation of cancer cells [99101]. For instance, blocking the Kv10.1 channel causes a cessation of cellular proliferation and migration in several myeloid leukemia cell lines [12].…”
