Gintonin, a novel ginseng-derived lysophosphatidic acid receptor ligand, improves brain functions and protects neurons from oxidative stress. However, little is known about the effects of gintonin against Pb-induced brain maldevelopment. We investigated the protective effects of gintonin on the developing cerebellum after prenatal and postnatal Pb exposure. Pregnant female rats were randomly divided into three groups: control, Pb (0.3% Pb acetate in drinking water), and Pb plus gintonin (100 mg/kg, p.o.). Blood Pb was increased in dams and pups; gintonin treatment significantly decreased blood Pb. On postnatal day 21, the number of degenerating Purkinje cells was remarkably increased while the number of calbindin-, GAD67-, NMDAR1-, LPAR1-immunoreactive intact Purkinje cells, and GABA transporter 1-immunoreactive pinceau structures were significantly reduced in Pb-exposed offspring. Following Pb exposure, gintonin ameliorated cerebellar degenerative effects, restored increased pro-apoptotic Bax, and decreased anti-apoptotic Bcl2. Gintonin treatment attenuated Pb-induced accumulation of oxidative stress (Nrf2 and Mn-SOD) and inflammation (IL-1β and TNFα,), restoring the decreased cerebellar BDNF and Sirt1. Gintonin ameliorated Pb-induced impairment of myelin basic protein-immunoreactive myelinated fibers of Purkinje cells. Gintonin attenuated Pb-induced locomotor dysfunctions. The present study revealed the ameliorating effects of gintonin against Pb, suggesting the potential use of gintonin as a preventive agent in Pb poisoning during pregnancy and lactation.

Background: Methyl mercury (Me Hg) is an environmental toxin associated with many serious neurological disorders. Conditioned Medium (CM) derived from Mesenchymal Stem Cells (MSCs) is a novel promising approach for the treatment of nervous system damage and various diseases. Objective: The aim of this research is to assess the consequence of Me Hg on the rats' cerebellar cortex and the potential neuroprotective effect of MSCs-CM. Materials and methods: Forty adult male albino rats were divided into four groups: Group I: control rats; Group II: Me Hg chloride treated rats; Group III: Me Hg chloride and DMEM treated rats; Group IV: CM treated rats after injection with Me Hg chloride. Cerebellar specimens were taken and handled for histological and immunohistochemical techniques. Morphometrical studies and statistical analysis were performed. Results: Groups II and III showed various changes such as neuronal degeneration and apoptosis. The mean number of Purkinje cells was significantly decreased(P<0.01), while Glial Fibrillary Acidic Protein (GFAP) immunostaining was significantly increased (P<0.01) in the neuroglial cells. Ultrastructural examination showed thinning and apparent decrease in the number of myelinated nerve axons. Shrunken Purkinje cells were observed with irregular nuclei, heterogenous cytoplasm, and disrupted mitochondria. Group IV showed improvement of the histological and electron microscopic changes defined in groups II and III. Conclusion: Me Hg exposure led to degenerative changes on cerebellar cortex. MSCs-CM is a very promising approach and has neuroprotective effects.

Introduction: Benign prostatic hyperplasia (BPH) is a common non-malignant overgrowth of human prostate in old age that greatly affects patient's quality of life. Finasteride, one of the routinely available regimens for BPH, caused several drawbacks. Recently a natural herbal product, ginseng, had shown a promising influence on various disorders through anti-proliferative, anti-inflammatory and other beneficial effects. Aim of the Work:To compare the effect of panax ginseng versus finasteride on BPH. Together with evaluation of the spontaneous improvement of hyperplastic features. Materials and Methods: Fifty five adult albino rats were divided into 2 groups: control and experimental groups. BPH was induced by subcutaneous injection of testosterone (3 mg/kg/day) for 4 weeks. Then the animals were subdivided equally into 4 subgroups: BPH was sacrificed at end of 4th week, Recovery was left untreated for another 4 weeks, Finasteride & Ginseng treated subgroups received oral administration of finasteride (5 mg/kg/day) & ginseng (200 mg/kg/day) respectively for another 4 weeks. Serum DHT level and weight of prostate glands were measured. Prostatic sections were stained with toluidine blue, H&E, Masson trichrome and immunohistochemical stain for PCNA and α SMA. Additionally, the sections were subjected to morphometric and statistical analysis. Results: BPH subgroup showed signs of hyperplasia of both epithelial and stromal cells while minimal improvement was demonstrated in the recovery subgroup. Finasteride treated subgroup showed apparent incomplete restoration of normal prostatic histological structure. While nearly normal histological architecture, biochemical & morphometric parameters were recorded in ginseng treated subgroup. Conclusion: Ginseng proved to have a therapeutic effect superior to finasteride through its anti-mitotic, anti-inflammatory and anti-fibrotic effects. Discontinuation of testosterone administration resulted in inconsiderable regression of BPH.
