Although the colonisation of coastal rivers on the Atlantic and Mediterranean coast by glass eels, Anguilla anguilla, has been well studied and understood, the colonisation of lagoons by glass eels is much less known. For the first time in the Mediterranean region, the installation of a glass eel fish-pass in Grau de la Fourcade channels in the Rhône delta enabled us to determine which factors could explain the variations in the catches of glass eel entering the Vaccarès coastal lagoon system. Whatever be the procedure chosen, the results of the model were the same: the temperature, the cumulative water discharge from the channel in the 5 nights before the catch (freshwater lure) and time that the drainage pumps were working explained the glass eel catches in the fish-pass in the Grau de la Fourcade. The tide and the cumulative discharge from the channel for only 3 nights before the catch did not seem to have a significant role in explaining catches. These results show that it is important that the lagoons should continue to receive rainfall runoff from their watersheds so that their water levels are high in winter, and that there is a good colonisation by glass eels as a result of a freshwater lure effect, when strong north winds expel low salinity water to the sea.

Exploration and Mining of the Bacterial Terpenome

CONSPECTUS Tens of thousands of terpenoids are present in both terrestrial and marine plants as well as fungi. In the last 5–10 years, however, it has become evident that terpenes are also produced by numerous bacteria, especially soil-dwelling Gram-positive organisms such as Streptomyces and other Actinomycetes. Although some microbial terpenes, such as geosmin—the degraded sesquiterpene responsible for the smell of moist soil, the characteristic odor of the earth itself—have been known for over 100 years, few terpenoids have been identified by classical structure- or activity-guided screening of bacterial culture extracts. In fact, the majority of cyclic terpenes from bacterial species have only recently been uncovered by the newly developed techniques of “genome mining.” In this new paradigm for biochemical discovery, bacterial genome sequences are first analyzed with powerful bioinformatic tools, such as the BLASTP program or Profile Hidden Markov models, to screen for and identify conserved protein sequences harboring a characteristic set of universally conserved functional domains typical of all terpene synthases. Of particular importance is the presence of variants of two universally conserved domains, the aspartate-rich DDXX(D/E) motif and the NSE/DTE triad, (N/D)DXX(S/T)XX(K/R)(D/E). Both domains have been implicated in the binding of the essential divalent cation, typically Mg2+, that is required for cyclization of the universal acyclic terpene precursors, such as farnesyl and geranyl diphosphate. The low level of overall sequence similarity among terpene synthases, however, has so far precluded any simple correlation of protein sequence with the structure of the cyclized terpene product. The actual biochemical function of a cryptic bacterial (or indeed any) terpene synthase must therefore be determined by direct experiment. Two common approaches are (i) incubation of the expressed recombinant protein with acyclic allylic diphosphate substrates and identification of the resultant terpene hydrocarbon or alcohol and (ii) in vivo expression in engineered bacterial hosts that can support the production of terpene metabolites. One of the most attractive features of the coordinated application of genome mining and biochemical characterization is that the discovery of natural products is directly coupled to the simultaneous discovery and exploitation of the responsible biosynthetic genes and enzymes. Bacterial genome mining has proved highly rewarding scientifically, already uncovering more than a dozen newly identified cyclic terpenes (many of them unique to bacteria) as well as several novel cyclization mechanisms. Moreover, bioinformatic analysis has identified more than 120 presumptive genes for bacterial terpene synthases that are now ripe for exploration. Fishes use selective tidal-stream transport (STST) during the first phase of this colonisation, and counter-current swimming during the second phase. Therefore, catadromous species have to achieve a behavioural shift, from STST to constant counter-current swimming. This has not yet been observed, and the location and period of this shift is still unknown. Our experimental protocol aimed to mimic the spatial progression of crossing the tidal limit within a 3-week experiment. Two catadromous fishes, thinlip mullets and European eels, were initially subjected to current reversal every 6.2 h during the first week. A gradual tidal distortion was performed during the second week, and fishes were submitted to a unidirectional water current during the third week. Our results reveal that all catadromous species use STST as far as possible within the tidal limit. At this point, in this experimental study, no young-of-the-year (YOY) fishes shifted from STST to constant counter-current swimming. This confirms that the behavioural shift occurs later, and that the second part of the upstream migration, counter-current progression, is performed by larger, older fishes and not YOY fishes. Les thalassotoques doivent donc réaliser une bascule comportementale entre ces deux comportements. Cette bascule n'a pas encore été observée directement. Sa localisation ainsi que sa période restent inconnues encore à ce jour. Notre protocole expérimental visait à reproduire en mésocosme la traversée de la limite tidale, lors d'une expérience longue de 3 semaines. Des individus de deux espèces thalassotoques, le mulet porc et l'anguille européenne, ont été initialement soumis à une renverse de courant toutes les 6,2 heures durant une semaine.
