Role of Fission Yeast Tup1-like Repressors and Prr1 Transcription Factor in Response to Salt Stress

Abstract: In Schizosaccharomyces pombe, the Sty1 mitogen-activated protein kinase and the Atf1 transcription factor control transcriptional induction in response to elevated salt concentrations. Herein, we demonstrate that two repressors, Tup11 and Tup12, and the Prr1 transcription factor also function in the response to salt shock. We find that deletion of both tup genes together results in hypersensitivity to elevated cation concentrations (K(+) and Ca(2+)) and we identify cta3(+), which encodes an intracellular catio… Show more

“…For example, heat-specific gene expression was independent of Sty1p, whereas H 2 O 2 -specific gene expression depended to a large part on Sty1p. The characterization of the cation-specific induction of cta3 in S. pombe illustrates how Sty1p may regulate stress-specific genes (Greenall et al, 2002). cta3 encodes a cation transporter, whose induction requires both activation of Sty1p and derepression of the Tup11/12p complex.…”

“…In addition to its Pap1-supporting role (Chen et al, 2008;Calvo et al, 2012), Prr1 induces stress genes independently of Sty1 (Ohmiya et al, 1999(Ohmiya et al, , 2000 and it supports the induction of Atf1-dependent genes (Calvo et al, 2012;Greenall et al, 2002). No orthologous protein of Prr1 exists in humans (Penkett et al, 2006), while one of the Prr1 orthologs in budding yeast, SKN7, assists the YAP1 TF (i.e.…”

“…Tup1 represses the expression of genes via distinct mechanisms: by establishing a repressive chromatin structure around the target gene promoter, by recruiting histone deacetylases, and by directly interfering with the general transcription machinery (14)(15)(16)(17)(18). Two Tup1 orthologs in Schizosaccharomyces pombe, Tup11 and Tup12 (Tup11/12), regulate multiple stress-responsive genes, including the fbp1 ϩ and cta3 ϩ genes, to provide stress specificity (19,20). However, the precise molecular mechanisms of Tup1 family proteins in gene repression have not been fully uncovered.…”
