Role of endothelin receptors in the hypertensive state of kinin B2 knockout mice subjected to a high-salt diet

Abstract: Mice with disruption of the kinin B(2) receptor (B(2)KO mice) are sensitive to salt-rich diets, which causes hypertension. The aim of the study was to assess the role of endothelin-1 (ET-1) and angiotensin-II in hypertensive B(2)KO mice on a salt-rich diet. We also wanted to verify if there is an upregulation of the mRNA expression of the precursors or receptors for these hormones. Two groups of B(2)KO mice (20-25 g) were investigated. The first group received an 8% NaCl diet with 1% NaCl in drinking water (HS… Show more

“…The concentration of 0.5% DMSO has been proved to be harmless in animals with i.p. injection [21,22]. Bosentan (Actelion Pharmaceuticals Japan Ltd, Tokyo, Japan) was prepared fresh each week as a suspension in 5% arabic gum and administered (100 mg?kg…”

“…Interestingly, we have recently shown that the BALB/c lungs present much lower levels of endothelin than C57Bl/6 under physiological conditions [15]. Thus, a low level of endothelins in BALB/c seems to be associated with high infiltration of lymphocytes.…”

“…BQ 788 (ET B receptor antagonist) was dissolved in PBS with 5% DMSO to obtain a solution of 1 mg/ml and used at 1 mg/kg. DMSO has to be used because these antagonists are insoluble in aqueous solution at this concentration according to Brochu et al [15]. The concentration of the antagonists was chosen from previous studies [16][17][18][19].…”

“…Null animals show increased sensitivity to deoxycorticosterone, with a more rapid and pronounced increase in blood pressure than controls (75), and increased sensitivity to high-salt diet hypertension, with renal blood flow reduced by 20%, and renal vascular resistance doubled compared with a normal-salt diet (9), indicating a tonic vasodilatory effect of bradykinin in this vascular bed. Salt-sensitive hypertension can be reversed by selective ET A and ET B antagonists, and also by inhibitors of ACE and AT 1 receptors, suggesting that ET-1 and ANG II are involved in the development of hypertension (39).…”
