Role of dynamin, synaptojanin, and endophilin in podocyte foot processes

Abstract: Podocytes are specialized cells that play an integral role in the renal glomerular filtration barrier via their foot processes. The foot processes form a highly organized structure, the disruption of which causes nephrotic syndrome. Interestingly, several similarities have been observed between mechanisms that govern podocyte organization and mechanisms that mediate neuronal synapse development. Dynamin, synaptojanin, and endophilin are functional partners in synaptic vesicle recycling via interconnected actio… Show more

“…There has been much speculation about the origin of these vesicles (36-39), but their functional significance has remained unresolved. Recent studies have demonstrated that impairment of the endocytic machinery in podocytes leads to proteinuria, indicating that endocytosis may play a crucial role in podocyte function (19,40,41). Our results reveal that podocytes induce significant levels of macropinocytosis in response to FFAs.…”

“…31 Neither the CD pdKO mouse podocytes nor the podocytes of their CD Ctrl littermates displayed significant colocalization of podocin when tested with an early endosomal marker Rab5 (Supplemental Figure 2B). By contrast, a late endosomal marker for Rab7 was increased in CD pdKO mouse podocytes compared with CD Ctrl littermate podocytes ( Figure 8B, Supplemental Figures 2C and 3B).…”

“…39 Glomeruli were harvested and enriched from mice kidney cortical tissue with 45% Percoll solution (GE Healthcare BioSciences, Uppsala, Sweden) as previously described. 40 The kidneys were harvested immediately and washed in ice-cold Krebs-Henseleit saline solution (119 mM NaCl, 4.7 mM KCl, 1.9 mM CaCl 2 , 1.2 mM L KH 2 PO 4 , 1.2 mM MgSO 4 , 7H 2 O, and 25 mM NaHCO 3 , pH, 7.4).…”
