Role of dopamine and selective dopamine receptor agonists on mouse ductus arteriosus tone and responsiveness

Abstract: Background: Indomethacin treatment for patent ductus arteriosus (PDA) is associated with acute kidney injury (AKI). Fenoldopam, a dopamine (DA) DA 1-like receptor agonist dilates the renal vasculature and may preserve renal function during indomethacin treatment. However, limited information exists on DA receptor-mediated signaling in the ductus and fenoldopam may prevent ductus closure given its vasodilatory nature. Methods: DA receptor expression in CD-1 mouse vessels was analyzed by qPCR and immunohistochem… Show more

“…Ductus and ascending aorta segments from 4 to 8 fetuses representing at least 4 litters were used for each myography study. Wnt1 Cre2 Prdm6 fl/fl and control vessels were freshly isolated, and vasoreactivity was evaluated using cannulated, pressurized vessel myography and computer-assisted videomicroscopy, as previously described (38)(39)(40)(41)61). Briefly, the excised vessel was mounted in custom myography chambers (University of Vermont), then equilibrated for 40 minutes at 37°C and 5 mmHg of distending pressure while submerged in modified, deoxygenated Krebs buffer.…”

“…Fetuses were delivered on D19 via terminal cesarean section, their thoracic cavities opened, and submerged in chilled and deoxygenated modified Kreb’s buffer, containing (in mM) 109 NaCl, 4.7 KCl, 2.5 CaCl 2 ·2H 2 O, 0.9 MgSO 4 , 1.0 KH 2 PO 4 , 11.1 glucose, and 34 NaHCO 3 (pH 7.3), as previously described ( 42 ). Fresh DA segments were excised, mounted in custom myography chambers (University of Vermont), and maintained at 37.5°C under deoxygenated conditions.…”
