Role of CD34 family members in lumen formation in the developing kidney

Abstract: Previous studies have shown CD34 family member Podocalyxin is required for epithelial lumen formation in vitro. We demonstrate that Endoglycan, a CD34 family member with homology to Podocalyxin, is produced prior to lumen formation in developing nephrons. Endoglycan localizes to Rab11-containing vesicles in nephron progenitors, and then relocalizes to the apical surface as progenitors epithelialize. Once an apical/luminal surface is formed, Endoglycan (and the actin-binding protein Ezrin) localize to large, in… Show more

“…As part of the CD34 family of transmembrane sialomucin proteins, its composition makes it both negatively charged and slippery due to glycosylation. Thus, Podocalyxin may help opposing apical membranes open by electrostatic repulsion and anti-adhesion 50,51 . In summary, the trafficking events mediating apical membrane establishment and lumen formation require an intricate and coordinated network of trafficking machinery, polarity complexes, and membrane proteins.…”

“…Nonparaffin sections were fixed in 4% PFA/PBS, permeabilized with 0.3% Triton X-100/PBS (PBST), and blocked with 10% donkey sera/PBST, as performed previously. 31 Sections were incubated with primary antibodies overnight (4°C), then with fluorophore-conjugated secondary antibodies, and mounted with Prolong Gold (Invitrogen). Immunohistochemistry was performed similarly except secondary antibodies were HRP-conjugated and DAB was used for staining.…”
