Role of CD1d- and MR1-Restricted T Cells in Asthma

Abstract: Innate T lymphocytes are a group of relatively recently identified T cells that are not involved in either innate or adaptive immunity. Unlike conventional T cells, most innate T lymphocytes express invariant T cell receptor to recognize exogenous non-peptide antigens presented by a family of non-polymorphic MHC class I-related molecules, such as CD1d and MHC-related molecule-1 (MR1). Invariant natural killer T (iNKT) cells and mucosal-associated invariant T (MAIT) cells quickly respond to the antigens bound t… Show more

“…In the allergy, the role of NKT cells is to maintain the Th1/Th2 balance by involving endo- and exogenous ligands for toll-like receptor 4 (TLR4) in iNKT cells [ 31 ]. There are reports showing the increased concentration of iNKT cells in bronchoalveolar lavage as well as in the peripheral blood in people with the exacerbation of bronchial asthma [ 32 , 33 ]. Similar observations were made in the case of atopic dermatitis.…”

“…The activated iNKT cells worsen the symptoms of allergic asthma independently of Th2‐cell‐mediated pathways. NKT cells are activated following antigenic stimulation in a CD1d‐dependent manner, as CD1d‐dependent antagonists or anti‐CD1d antibodies suppress OVA‐induced airway inflammation and histamine release 60 . The iNKT cells produce various cytokines such as IL‐4, IL‐5, IL‐13 and IL‐17 61 .…”

“…A role for less abundant T cell types in asthma is evolving (Table ). Both mouse and human studies have implicated invariant NKT cells (iNKT) in the pathogenesis of allergic asthma . These cells express T cell receptors (TCR) of limited diversity that recognize lipid antigens presented by the “non‐classical” MHC molecule, CD1d .…”

“…However, they comprise less than 2% of T cells in bronchoalveolar lavage (BAL) fluid from children with severe asthma . A similar cell type, mucosal‐associated invariant T (MAIT) cells, is abundant in the human lungs, and also displays limited TCR diversity . These cells respond to microbial‐derived vitamin B2 metabolites presented by the MHC class I‐related molecule MR1 and exert anti‐microbial effects by their cytotoxic activity and secretion of IFN‐γ and IL‐17 .…”
