Role of ARK5 in cancer and other diseases (Review)

Abstract: Malignant tumors are often exposed to hypoxic and glucose-starved microenvironments. AMP-activated protein kinase (AMPK) is an energy sensor that is stimulated during energy-deficient conditions and protects cells from hypoxic injury by regulating metabolism. AMPK-related protein kinase 5 (ARK5) is a member of the catalytic sub-unit of the AMPK family and has an important role in energy regulation and hypoxia. ARK5 is regulated by Akt and liver kinase B1 and is associated with numerous tumor-related molecules … Show more

“…ARK5 is upregulated by MAF and MAFB through the MARE sequence in MM cells [ 58 ]. In several cancers, overexpression of ARK5 is related to tumor invasion, metastasis, and poor prognosis, with downregulation of ARK5 resulting in improved sensitivity to anti-tumor drugs [ 60 ]; thus, ARK5 is a potential therapeutic target in MM with t(14;16) or t(14;20).…”

“…Previous studies have shown that ARK5 is involved in the regulation of EMT processes in a variety of tumors [ 14 ]. Therefore, we speculate that ARK5 has a regulatory role in hepatocyte EMT during liver fibrosis.…”

“…ARK5 is a serine/threonine kinase and has been identified as an essential part of the catalytic subunit of the AMPK (AMP-activated protein kinase) family [ 14 ]. Previous studies have found that ARK5 participates in a number of significant functions such as cell adhesion, cell proliferation and metastasis, and carbohydrate metabolism [ 15 ].…”

“…Despite the limited number of studies that experimentally elucidate the role of NUAK1 and NUAK2 in cancer development, both kinases have been found amplified in different tumors. The role of NUAK1 in different tumor types has been recently reviewed in [38]. Here, we highlight the role of NUAK proteins in different pathways and their potential involvement in tumor initiation and propagation.…”
