Role of AMPK/mTOR-independent autophagy in clear cell renal cell carcinoma

Abstract: We examined the status and role of autophagy, a process of lysosomal recycling of cellular material, in clear cell renal cell carcinoma (ccRCC). Paired samples of tumor and adjacent non-malignant tissue were collected from 20 patients with ccRCC after radical nephrectomy. The mRNA levels of apoptosis (BAD, BAX, BCL2, BCLXL, BIM) and autophagy (ATG4, BECN1, GABARAP, p62, UVRAG) regulators were measured by RT-qPCR. The protein levels of autophagosome-associated LC3-II, autophagy receptor p62, apoptotic marker PA… Show more

“…(26) Moreover, PI3K/Akt/mTOR pathway is defined as the most important pathway in regulation, initiation and progression of autophagy in RCC (27). By examining the role of autophagy and its signaling pathway in ccRCC, transcriptional induction of autophagy in ccRCC was found, accompanied by AMPK/mTOR-independent increase in ULK1 activation and autophagic flux, which might slow tumor progression and metastasis independently of apoptosis (28).…”

“…We noted that in addition to promoting proliferation, migration, and EMT, SPAG9 also promotes autophagy in several tumor-cell lines [ 18 , 19 ]. In ccRCC, increased autophagic flux was reported to be associated with better prognoses [ 35 , 36 ]; therefore, we speculated that SPAG9 participates in autophagy in ccRCC.…”

“…HIF-1α in renal cell carcinoma inhibits the activity of CPT1 on the outer mitochondrial membrane and prevents the β-oxidation of fatty acids ( 398 ), which is important for the differentiation of Treg cells, and its blockade could prevent the accumulation of this immunosuppressive population ( 199 ). AMPK in renal cancer cells, a sensor of nutrient deprivation and metabolic stress, is inactivated in the AMPK-GATA3-ECHS1 signaling pathway ( 399 ), inhibiting the expression of the transcription factor GATA3 and leading to a decrease in the synthesis of ECHS1. AMPK activation can promote the formation of Treg cells while reducing Th1 and Th17 cells ( 199 ), thus, leading to unwanted immune modulation in the context of RCC.…”
