Role of AMP‐Activated Protein Kinase in Cancer Therapy

Abstract: Recent advances in AMP-activated protein kinase (AMPK) as a target in cancer waxed and waned over the past decade of cancer research. AMPK is a cellular energy sensor, present in almost all eukaryotic cells. An elevated AMP/ATP ratio activates the AMPK, which in turn inhibits energy-consuming processes and induces catabolic events that generate ATP to restore the energy homeostasis inside the cell. Several reports have indicated that AMPK regulates several metabolic pathways and may be a potential therapeutic … Show more

“…For example, therapeutic activators of AMPK have been utilised in a number of diseases to counteract the inflammatory response [59]. AICAR treatment in murine models of experimental encephalomyelitis and asthma reduced pro-inflammatory cytokine production and injury [60,61].…”

“…To date, a number of studies have explained the role of AMPK in tumorigenesis [34]. Initially, the connection between AMPK and cancer biology was through the discovery of tumor suppressor LKB1 as a major AMPK upstream kinase [35].…”

“…In response to elevated AMP/ATP ratios, AMPK binds AMP and is then phosphorylated by LKB1. This leads to activated AMPK, suppression of anabolic metabolism, and a concomitant shift to catabolic events that generate ATP to restore energy homeostasis (2,10).…”
