Role of AGC kinases in plant growth and stress responses

Abstract: AGC kinases are important regulators of cell growth, metabolism, division, and survival in mammalian systems. Mutation or deregulation of members of this family of protein kinases contribute to the pathogenesis of many human diseases, including cancer and diabetes. Although AGC kinases are conserved in the plant kingdom, little is known about their molecular functions and targets. Some of the best-studied plant AGC kinases mediate auxin signaling and are thereby involved in the regulation of growth and morphog… Show more

“…Because OXI1 also is involved in root-hair growth and can phosphorylate different members of the RLCK-VIII subfamily, OXI1 is likely to activate MRI during root-hair growth. In PTs, however, OXI1 expression levels are very low, and it seems more likely that either another member of this family, the AGC2 kinases (group VIII), or a combination of them fulfills this role (Table S5) (35)(36)(37). Whether CrRLK1Ls, NADPH oxidases, OXI1/ AGC2 kinases, and MRI/Pti1-like proteins are all part of a linear pathway or, more likely, belong to different pathways that orchestrate complex cross-talk to sustain cell growth remains to be addressed.…”

“…Whether CrRLK1Ls, NADPH oxidases, OXI1/ AGC2 kinases, and MRI/Pti1-like proteins are all part of a linear pathway or, more likely, belong to different pathways that orchestrate complex cross-talk to sustain cell growth remains to be addressed. In this regard, the presence of a module composed of OXI/AGC2 kinases and MRI/Pti1-like RLCKs acting downstream of the CrRLK1Ls and NADPH oxidases could potentially allow the integration of inputs generated by biotic or abiotic stresses with the CW integrity pathway (36,37). Interestingly, in all the different genetic backgrounds tested, MRI R240C is more efficient than the WT form in activating downstream responses.…”

“…In mammalian systems, AGC kinases are important regulators of cell growth, metabolism, and cell death (Pearce et al, 2010). Although the catalytic kinase domains of plant AGC kinases share sequence similarities with those of mammalian AGC kinases, their molecular functions and targets are largely unknown (Garcia et al, 2012). In the Arabidopsis genome, there are 39 AGC kinase members (Bögre et al, 2003).…”

“…In the Arabidopsis genome, there are 39 AGC kinase members (Bögre et al, 2003). To date, in different model plant species, several AGC kinases have been implicated in various developmental and physiological processes, including growth, morphogenesis, defense, and symbiosis (Rentel et al, 2004;Abuqamar et al, 2008;Fu et al, 2009;Garcia et al, 2012;Rademacher and Offringa, 2012;Zhu et al, 2015). In Arabidopsis, OXI1 is required for immunity against the pathogenic oomycete Hyaloperonospora arabidopsidis and the bacterial biotrophic pathogen Pseudomonas syringae pv tomato DC3000 (Rentel et al, 2004;Petersen et al, 2009).…”

“…Moreover, defects in the INCOMPLETE ROOT HAIR ELONGATION (IRE) gene, which encodes another AGC protein kinase, caused a similar short-root-hair phenotype to agc2-1 mutants (Oyama et al, 2002). These findings, together with the existence of numerous AGC kinases in plants (Garcia et al, 2012), suggest that AGC protein kinase cascades integrate various signals including phospholipids and ROS in the regulation of root hair tip growth. It is noteworthy that PDK1 phosphorylates PIONOID (PID), the AGC protein kinase that modulates auxin polar transport via the phosphorylation of PINFORMED (PIN) auxin transporters (Zegzouti et al, 2006;Huang et al, 2010).…”
