RNP transport in cell biology: the long and winding road

Abstract: Regulation of gene expression is key determinant to cell structure and function. RNA localization, where specific mRNAs are transported to subcellular regions and then translated, is highly conserved in eukaryotes ranging from yeast to extremely specialized and polarized cells such as neurons. Messenger RNA and associated proteins (mRNP) move from the site of transcription in the nucleus to their final destination in the cytoplasm both passively through diffusion and actively via directed transport. Dysfunctio… Show more

“…We next set out to determine the molecular mechanism of RP-mRNAs localization to protrusions. Localization of mRNAs is driven by specific RBPs which bind to and mediate transport or subcellular anchoring of their target transcripts (Eliscovich and Singer, 2017). We 145 therefore hypothesized that specific protrusion localized RBPs must be interacting with and localizing RP-mRNAs to protrusions.…”

“…Interestingly, different cell types such as neurons with protrusions exceeding hundreds of µm, and compact cell types such as fibroblasts, utilize different mechanisms to distribute β-actin mRNA 20 . The diversity of mRNA dynamics highlights the need to investigate mRNA movement in different cell types and interrogate the complex network of mRNA binding proteins and their role for distributing mRNAs within cells 21 .…”

“…Studies from several model transcripts have uncovered general principles that dictate how mRNAs become localized within cells. Conserved cis ‐acting sequences, usually within the mRNA 3′‐untranslated region (3′UTR), are recognized by trans ‐acting RNA‐binding proteins (RBPs) to regulate mRNA transport, translation, stability and degradation . Each RBP can interact with few or many target mRNAs on the basis of primary sequence and/or secondary structure.…”

“…RNA-binding proteins (RBPs) to regulate mRNA transport, translation, stability and degradation. 2 Each RBP can interact with few or many target mRNAs on the basis of primary sequence and/or secondary structure. Most localized mRNAs interact with multiple RBPs throughout their lifetime, and the localization of multiple mRNAs to the same subcellular domain is often regulated by the same RBPs.…”
