RNASeq in C. elegans Following Manganese Exposure

Abstract: Manganese is a metal that is required for optimal biological functioning of organisms. Absorption, cellular import and export, and excretion of manganese are all tightly regulated. While some genes involved in regulation, such as DMT-1 and ferroportin are known, it is presumed that many more are involved and as yet unknown. Excessive exposure to manganese, usually in industrial settings, such as mining or welding, can lead to neurotoxicity and a condition known as manganism that closely resembles Parkinson's d… Show more

“…In combination, these assays are even more powerful. For example, Parmalee and colleagues have characterized a robust method to perform RNA-Seq in C. elegans exposed to manganese, which could be easily adaptable for any environmental exposure [ 73 ]. Combined with the relative ease of genetic manipulation in C. elegans , platforms such as these are ideal for amassing gene-environment information relative to disease mechanisms in PD, like mitochondria [ 74, 75 ] and monoamine function [ 76 ].…”

“…The J1-stage worms can be transferred to seeded plates to continue development until the desired stage. The synchronization process summarized below has been adapted from previously described methods (e.g., Lewis & Fleming, 1995; see Current Protocols article; Parmalee et al, 2015). 2.…”

“…In order to understand the overall toxic effects of DON in C. elegans , we employed RNAseq to analyze the comprehensive modulation of gene expression in C. elegans upon DON intoxication. RNAseq analysis has been used to elucidate transcriptional changes following manganese exposure in C. elegans [ 14 ]. Whole genome microarray analysis has similarly been used to profile the host response in mouse brain following exposure to the plant toxin abrin [ 15 ].…”
