RNAi-mediated knockdown of gut receptor-like genes prohibitin and α-amylase altered the susceptibility of Galleria mellonella to Cry1AcF toxin

Abstract: Background Due to the prolonged usage of Bt-based biopesticides and Bt-transgenic crops worldwide, insects are continually developing resistance against Cry toxins. This resistance may occur if any mechanistic step in the insecticidal process is disrupted possibly because of the alteration in Cry-receptor binding affinity due to mutation in receptor genes. Compared to other lepidopteran insects, Cry receptor-related research has made asymmetric progress in the model insect Galleria mellonella. … Show more

“…qPCRbased expression analysis of various candidate genes (primer details given in supplementary Table 1) was performed in a CFX96 thermal cycler (BioRad). The PCR efficiency of each primer pair was calculated by using different primer concentrations in different RT-qPCR reactions, followed by generating the standard curve [53]. 10 µL of RT-qPCR reaction volume constituted 1.5 ng cDNA, 750 nM each of sense and antisense primers, and 5 µL SYBR Green PCR master-mix (BioRad).…”
