RNA sequencing reveals an additional Crz1-binding motif in promoters of its target genes in the human fungal pathogen Candida albicans

Abstract: Background: The calcium/calcineurin signaling pathway is mediated by the transcription factors NFAT (nuclear factor of activated T cells) in mammals and Crz1 (calcineurin-responsive zinc finger 1) in yeasts and other lower eukaryotes. A previous microarray analysis identified a putative Crz1-binding motif in promoters of its target genes in Candida albicans, but it has not been experimentally demonstrated. Methods: An inactivation mutant for CaCRZ1 was generated through CRISPR/Cas9 approach. Transcript profili… Show more

“…Regardless of the patterns of Dp or Dr changes, this elevated FA in the grey matter post-TBI has been associated with astrogliosis [ 25 , 27 ] or microstructural remodelling in a rat model of open-head injury [ 95 ]. Of particular interest is the association of DTI/NODDI changes and astrocyte activity, since astrocytes have dual roles in neuronal plasticity and reconstruction after traumatic brain injury [ 96 , 97 ].…”

“…Further studies are required to warrant any conclusion. Moreover, recent report demonstrates that microRNA-941 protects endometrial cells from oxygen and glucose deprivation-re-oxygenation by activation of Nrf2 signalling through targeting Keap1, a Nrf2 suppressor protein 79 . Nrf2 overexpression promotes cancer progression and drug resistance in various cancers by activating several oncogenes such as matrix metallopeptidase 9, B-cell lymphoma 2, tumour necrosis factor α, and vascular endothelial growth factor A 80 82 .…”

“…Defects in calcium homeostasis would lead to the retention of cytoplasmic calcium, which in turn activates calcineurin phosphatase activity [ 27 ]. The activated calcineurin will dephosphorylate zinc finger transcription factor Crz1, promoting nuclear translocation and induction of genes involved in azole resistance in Candida albicans [ 31 ]. Although the relationship between the calcineurin–Crz1 pathway and azole resistance has been determined, it is still unclear whether there is a direct relationship between the calcium signaling pathway and mitochondrial function.…”
