RNA-Seq profiling of circular RNAs in human colorectal Cancer liver metastasis and the potential biomarkers

Circular RNAs (circRNAs) make up a large class of non‐coding RNAs and play important roles in the pathology of a variety of diseases. However, their roles in pulmonary macrophage polarization after sepsisinduced lung injury is unknown. In this study, mice were divided into two groups: Sham control group and cecal ligation and puncture (CLP)‐induced ALI group. Macrophages were isolated from lung homogenates 24 hours after SCLP/CLP. We started with RNA‐seq of circRNA changes in macrophages and validated by RT‐PCR in the following experiments. A total of 4318 circRNAs were detected in the two groups. Of these, 11 and 126 circRNAs were found to be significantly upregulated and downregulated, respectively, compared to the control (p≤0.05, Fold Change ≥2). Differentially expressed circRNAs with a high foldchange (fold‐change >4, P<0.05) were selected for validation by qRT‐PCR, 10 of which were verified. Furthermore, the most differentially expressed circRNAs within all the comparisons were annotated in detail with circRNA/miRNA interaction information using miRNA target prediction software. The network of circRNA‐miRNA‐mRNA was illustrated by cytoscape software. Gene ontology analyses indicated the upregulated circRNAs were involved in the multiple biological functions such as regulation of mitochondrion distribution and Notch binding, while the down‐regulated circRNAs mainly involved in the biological process as histone H3K27 methylation. KEGG pathway analysis revealed TGF‐beta signaling pathway was related to the upregulated circRNAs. The present study provides a novel insight into the roles of circRNAs in pulmonary macrophage differentiation and polarization post septic lung injury.

Roles of circRNAs in the tumour microenvironment

The tumour microenvironment (TME) constitutes the area surrounding the tumour during its development and has been demonstrated to play roles in cancer-related diseases through crosstalk with tumour cells. Circular RNAs (circRNAs) are a subpopulation of endogenous noncoding RNAs (ncRNAs) that are ubiquitously expressed in eukaryotes and have multiple biological functions in the regulation of cancer onset and progression. An increasing number of studies have shown that circRNAs participate in the multifaceted biological regulation of the TME. However, details on the mechanisms involved have remained elusive until now. In this review, we analyse the effects of circRNAs on the TME from various perspectives, including immune surveillance, angiogenesis, hypoxia, matrix remodelling, exo-circRNAs and chemoradiation resistance. Currently, the enormous potential for circRNA use in targeted therapy and as noninvasive biomarkers have drawn our attention. We emphasize the prospect of targeting circRNAs as an essential strategy to regulate TME, overcome cancer resistance and improve therapeutic outcomes.

Purpose: As a novel type of non-coding RNAs, circRNAs were found to play important roles in cancer progression. In this study, we aim to investigate genome-wide circRNAs expression and potential functions in glioma to find new therapeutic targets for glioma treatment. Patients and Methods: A total of 92 pairs of glioma tissue samples and adjacent control samples were enrolled in this study. Microarray and bioinformatics tools were used to identify circRNAs expression in glioma. Relative expression of RNAs was validated by qRT-PCR. Cell viability and migration were measured by Cell Counting Kit-8 and wound-healing assay in U251 and SHG-44 cells. The statistical correlation and overall survival were calculated by Mann-Whitney U-test and Kaplan-Meier method. Results: A total of 90 differentially expressed circRNAs were identified in glioma tissues compared to control. These circRNAs were mainly back-spliced forms chr1, chr6 and chr12 with a variable number of exons. QRT-PCR showed hsa_circ_0013520 and hsa_circ_0004379 were highly expressed in glioma tissues and cell lines. High expression of hsa_circ_0013520 and hsa_circ_0004379 was significantly correlated with tumor size (P-value < 0.001), Karnofsky Performance Status (KPS, P-value: 0.008 and 0.006) and TNM stage (P-value: 0.004 and 0.005). Moreover, patients with higher expression level of hsa_circ_0013520 and hsa_circ_0004379 had a poorer overall survival (P-value: < 0.01). Besides, loss-of-function experiments revealed that inhibition of hsa_circ_0013520 and hsa_circ_0004379 suppresses proliferation and invasion of glioma cells. Conclusion: The present study is the first to report that hsa_circ_0013520 and hsa_-circ_0004379 were up-regulated in glioma tissues and cell lines, and their expression was positively correlated with poor clinical features of glioma patients, which may serve as novel biomarkers for glioma diagnosis and therapy.
