RNA-seq analysis reveals TRPC genes to impact an unexpected number of metabolic and regulatory pathways

Abstract: The seven-member transient receptor potential canonical genes (TRPC1-7) encode cation channels linked to several human diseases. there is little understanding of the participation of each tRpc in each pathology, considering functional redundancy. Also, most of the inhibitors available are not specific. Thus, we developed mice that lack all of the TRPCs and performed a transcriptome analysis in eight tissues. the aim of this research was to address the impact of the absence of all tRpc channels on gene expressi… Show more

“…They are therefore able to mediate cellular responses to these signals and interact with important hippocampal functions. and manganese ion transport were modified in both forebrain and midbrain [50]. TRPC channels are involved in a number of mechanosensory processes [51,52]; however, recent papers seem to point out that they are more likely activated by mechanosensitive processes than directly mechanosensitive [53,54].…”

“…All animal procedures were carried out in accordance with the NIH guidelines for the Care and Use of Laboratory Animals and approved by the Animal Welfare Committee of the University of Texas Health Science Center at Houston. TRPC1 KO ( Trpc1 –/– ) and quadruple TRPC1, C4, C5, C6 KO (QuadKO) mice (129/SvEv and C57BL/6J mixed background) were created and maintained as described ( Dietrich et al, 2007 ; Formoso et al, 2020 ); TRPC4 KO ( Trpc4 –/– ) in C57BL/6 background were generated and maintained as previously described ( Freichel et al, 2001 ).…”

“…Recent data showed the possible role of transient receptor potential canonical (TRPC) genes by performing RNAseq analysis in mice that lack the TRPC [46]. The authors pointed out the deregulation of TRPC belonging to several metabolic pathways such as PI3K-B signaling pathway, cytokine-cytokine receptor interaction, calcium homeostasis, extracellular matrix -receptor, and circadian rhythms in several human diseases.…”
