RNA Interference of Odorant-Binding Protein 2 (OBP2) of the Cotton Aphid, Aphis gossypii (Glover), Resulted in Altered Electrophysiological Responses

Abstract: Aphis gossypii (Glover) (Hemiptera: Aphididae) is a highly invasive pest that feeds primarily on phloem resulting in severe economic loss to growers. A. gossypii has cosmopolitan distribution with broad host range, polyphenism, parthenogenetic mode of reproduction, vectoring abilities, and host alteration which has profound influence on its management. Odorant-binding proteins (OBPs) in insects are involved in olfaction, playing a key role in orienting the insect for feeding or oviposition. Recent studies reve… Show more

“…We then surmised that silencing OBPs might lead to behavioral changes in the phenotype. After all, gene silencing has already been successfully applied to investigate functions of OBPs in other insects (Chen et al, 2008; Pelletier et al, 2010; Rebijith et al, 2016). Of the two OBPs specific to male antennae, we selected RproOBP27 for these studies.…”

“…Several insects can nevertheless exhibit sustained RNAi for prolonged periods (Paim et al., ; Coleman et al., ; Khajuria et al., ), suggesting a system of signal amplification. Hemipterans appear to have a robust RNAi amplification system, as gene silencing has been demonstrated for 4 days (Rebijith et al., ), 6 days (Coleman et al., ), and 8 days (Tzin et al., ) after feeding with dsRNA. Coleman et al.…”

“…Similarly, Over-expression of Cytochrome P450 monooxygenase gene CYP6DA2 increased sensitivity to spirotetramat, alpha-cypermethrin and the toxicity of gossypol to cotton aphids [80,81]. Another study has demonstrated the silencing of the odorant-binding protein 2 ( OBP2 ) which resulted in impaired host-seeking and oviposition behavior of A. gossypii [82]. Furthermore, cotton transgenic plants expressing Amaranthus caudatus agglutinin (amaranthin) conferred enhanced resistance to A. gossypii [83].…”
