RNA-dependent disassembly of nuclear bodies

Abstract: Nuclear bodies are membraneless organelles that play important roles in genome functioning. A specific type of nuclear bodies known as interphase prenucleolar bodies (iPNBs) are formed in the nucleoplasm after hypotonic stress from partially disassembled nucleoli. iPNBs are then disassembled, and the nucleoli are reformed simultaneously. Here, we show that diffusion of B23 molecules (also known as nucleophosmin, NPM1) from iPNBs, but not fusion of iPNBs with the nucleoli, contributes to the transfer of B23 fro… Show more

“…The formation of interphase prenucleolar bodies (iPNBs) associated with unprocessed rRNA is induced in cells under hypotonic stress ( 74 ). Here, we show that the inhibition of RNA Pol I by Act D treatment induces the aggregation of nucleolin speckles in the nucleoplasm, which is further enhanced upon lamin B2 depletion ( Fig.…”

“…The optimal time for the start of iPNB imaging is 25-35 min after the return to isotonic conditions. At this stage, the number of iPNBs is substantially reduced [7], facilitating the imaging and analysis of iPNBs tracks. Cells can be incubated directly on the microscope stage, if the thermostatic chamber allows to control both CO 2 and temperature.…”

“…iPNBs are formed in interphase nuclei from partially disassembled nucleoli when cells return to isotonic conditions from a hypotonic shock [4]. iPNBs contain nucleolar proteins NPM1 (B23 or nucleophosmin) [4][5][6][7], NCL (C23 or nucleolin) [8] and pre-rRNAs [7]. In contrast to the majority of other NBs, a large number (several hundred per nucleus) of iPNBs is induced simultaneously, and they are uniformly distributed within the nuclear space [7].…”

“…On the other hand, maintenance of high RNA concentration prevents the formation of insoluble RNP complexes [6]. Studies of the nucleolus reassembly after stress suggest that RNA species, which initiates nuclear domain formation also drives transfer of proteins from disassembling transient nuclear bodies to its parental reassembling nuclear domain [7].…”
