Rift Valley Fever Virus L Protein Forms a Biologically Active Oligomer

Abstract: Rift Valley fever virus (RVFV) (genus Rift Valley Fever virus (RVFV), which belongs to the genusPhlebovirus in the family Bunyaviridae, is endemic in subSaharan African countries and causes large outbreaks in endemic areas and countries outside of the endemic area, including Egypt, Saudi Arabia, and Yemen (2). RVFV is transmitted by mosquitoes, and the virus infection causes a high rate of abortions in pregnant ruminants and acute lethal hepatitis in newborn lambs (26). Most human patients show an acute febril… Show more

“…R ift Valley fever virus (RVFV) is a member of the Phlebovirus genus of the Bunyaviridae family of viruses (1,2). It has a tripartite negative-stranded RNA genome consisting of small (S), medium (M), and large (L) segments encoding the N, NSs (3), Gn (G2), Gc (G1), NSm (4,5), and L genes, respectively (6,7). RVFV is spread primarily by infected Aedes mosquitoes and is the causative agent of Rift Valley fever (RVF), originally described following an outbreak of enzootic hepatitis on a farm in the Rift Valley of Kenya in 1931 (8).…”

“…The advantage of this method is its capacity to directly visualize, under physiological conditions, weak or transient protein-protein interactions that cannot be detected by traditional biochemical techniques. Although developed only recently, this assay has been widely employed to investigate viral protein-protein interactions (2,3,5,6,20,24,26,64). Previous reports using the GST pulldown assay have shown that amino acids 30 to 37 are essential for N-N interactions (59).…”
