Ribosomal RNA genes in B chromosomes of Crepis capillaris detected by non-radioactive in situ hybridization

Abstract: A non-radioactive in Situ hybridization method using biotin-labelled rDNA has made it possible to localize rRNA genes not only at the secondary constriction in both homologous chromosomes No. 3 of Crepis capillaris but also in the B chromosomes occurring in the plants employed. Very clear dot-like rDNA signals at the telomeres of both arms were observed in all B chromosomes. Histochemical silver staining, which is indicative of transcriptional activity of rRNA gene clusters, resulted in both darkly-staining nu… Show more

“…Second, rDNA is subject to very rapid copy number evolution between species [53][54][55] and within species [56][57][58]. It can extrachromosomally circularize [59,60], attract transposable elements [58,61] as well as invade other selfish elements, such as B chromosomes [62][63][64] and germline-limited DNA [65]. Finally, rRNA gene clusters are recombinational hotspots in cancer [66], and abnormalities in the nucleolar morphology of cancer cells attracted the attention of tumour pathologists as early as the nineteenth century [67,68].…”

“…These elements are potentially B-chromosomes. The fact that in some well-studied cases it has been possible to demonstrate the existence of transcription involving nucleolar organizer regions (Cabrero et a!., 1987;Maluszynska & Schweizer, 1989) indicates an ancestral relationship with the A chromosomes. The results obtained in E. plorans also suggest this ancestral resemblance.…”

“…The plants used were descendants of original plant and seed material collected from three areas near Port Augusta in South Australia during the spring of 1989and 1990(John et a!., 1991. Brachycome dichromosomatica is an outbreeding species and abundant seed can readily be obtained by rubbing flowering heads together.…”

“…The method of Maluszynska & Schweizer (1989) was modified as follows. Slides were incubated for 10 mm in 0.01 M borate buffer (pH 9.2), 200 1uL 1: 1 w/v AgNO3 in water was Millipore filtered and added to each slide, which was covered with a nylon mesh and incubated at 65°C for up to 2 h. Slides were rinsed, 200 uL 3:2 AgNO3: 2 per cent gelatine 1 per cent formic acid (pH 3.0) added and incubated for 10 mm.…”
